In this paper the effects on the interaction of highly positively charged substitution-inert platinum polynuclear complexes (SI-PPCs) with negatively charged DNA and heparin are examined and compared by theoretical chemistry methods. Electrostatic and hydrogen bonding interactions contribute to the overall effects on the biomolecule. Root Mean Square (RMS) deviation, Solvent Accessible Surface, RMS fluctuation, and interaction analysis all confirm similar effects on both biomolecules, dictated predominantly by the total positive charge and total number of hydrogen bonds formed. Especially, changes in structural parameters suggesting condensation and reduction of available surface area will reduce or prevent normal protein recognition and may thus potentially inhibit biological mechanisms related to apoptosis (DNA) or reduced vascularization viability (HEP). Thermodynamic analyses supported these findings with favourable interaction energies. The comparison of DNA and heparin confirms the general intersectionality between the two biomolecules and confirms the intrinsic dual-nature function of this chemotype. The distinction between the two-limiting mode of actions (HS or DNA-centred) could reflect an intriguing balance between extracellular (GAG) and intracellular (DNA) binding and affinities. The results underline the need to fully understand GAG-small molecule interactions and their contribution to drug pharmacology and related therapeutic modalities. This report contributes to that understanding.

Quantum mechanical approaches and molecular docking analysis of platinum metal-based anticancer drugs Lobaplatin and Heptaplatin targeting cancer DNA - a comparative analysis

The structural anal. and chem. reactivity of Lobaplatin and Heptaplatin were done using Spectral anal., NBO anal., and Global Chem. Reactivity and HOMO-LUMO values obtained using D. functional theory(DFT).Mol. Docking studies were performed for the drug compounds with DNA structure, Lung cancer, Gastric cancer, and Ovarian cancer cell structures.The obtained binding affinity results were compared and can be concluded that Hepataplatin shows the Lowest binding energy values with DNA structure followed by Lobaplatin than cisplatin and Lobaplatin shows good docking affinity towards Cancer cell structure comparatively.Lobaplatin and Heptaplatin were found to be highly interactive anticancer drug compounds

A review.Despite multiple advances in prevention and treatment, cancer remains a deadly disease globally.From 1942 to the present day, chemotherapy has been recognized as one of the main alternatives in the treatment of cancer.Nevertheless, the use of anticancer metallodrugs was investigated until 1965, when the therapeutic application of cisplatin was discovered circumstantially.Currently, other metallodrugs based on platinum and ruthenium have reached intermediate and advanced clin. research phases, some even reaching the market (carboplatin, heptaplatin, lobaplatin, miriplatin, nedaplatin, oxaliplatin, etc.).During the last decades, scientists have developed new metallodrugs that are more effective and selective, but with fewer side effects.Metal complexes based on transition metals have demonstrated very good anticancer activity, mainly when coordinated with organic ligands containing nitrogen or oxygen.In this sense, various reports have confirmed that Schiff bases derived from salicylaldehyde form ideal N₂O₂ ligands (salphen, salen, salpn) to design coordination complexes with d-block metals potentially useful in chemotherapy.Therefore, this document could pave the way for the scientific community focused on the design of new metallodrugs based on tetradentate Schiff bases and transition metals.
