This is a phase Ib/II clinical study that has two phases. In phase Ib, the safety evaluation of the extract of Petiveria alliacea (Esperanza) will be carried out in patients with metastatic gastrointestinal tumors (colon, pancreas, stomach, and biliary tract) and patients with newly diagnosed and relapsed acute leukemia. In phase IIb, the safety will continue to be evaluated, and the efficacy of the Esperanza extract will be explored in combination with chemotherapy in patients with metastatic gastrointestinal tumors (colon, pancreas, stomach, and biliary tract) with newly diagnosed acute leukemias and relapses.

Study Characterizing the Impact of Different Therapeutic Strategies on Event Occurrence at 2 Years, 5 Years, 10 Years, and 15 Years, According to Prognostic Groups in Patients With Hodgkin Lymphoma

RATIONALE: Drugs used in chemotherapy work in different ways to stop the growth of cancer cells, either by killing the cells or by stopping them from dividing. Giving drugs in different combinations may kill more cancer cells. It is not yet know which treatment regimen is more effective in treating Hodgkin lymphoma.
PURPOSE: This phase III trial is studying three different therapy regimens to compare how well they work in treating patients with previously untreated Hodgkin lymphoma.

Methanolic extract of wheatgrass (Triticum aestivum L.) prevents BPA-induced disruptions in the ovarian steroidogenic pathway and alleviates uterine inflammation in Wistar rats

The present study examined the anti-inflammatory and functional improvement of the uterus and ovary, respectively, in bisphenol-A (BPA)-fed adult Wistar rats following the ingestion of methanolic extract of wheatgrass (WG-ME). Four groups of rats were conditioned as vehicle-treated control, BPA-treated (100 mg/kg b.w.), BPA + WG-ME (100 mg BPA/kg b.w. + 200 mg WG-ME/kg b.w.), and WG-ME (200 mg/kg b.w.) groups. The LC-MS study confirmed the presence of numerous bioactive components in WG-ME. ELISA, PAGE, real-time PCR, and immunohistostaining were executed to test the efficacy of WG-ME against BPA. WG-ME was shown to induce significant weight gain of the uterus and ovaries as well as improve the estrous cycle and antioxidant status. WG-ME effectively suppressed the mRNA expression of TNF-α (tumor necrosis factor-alpha) and NF-κB (nuclear factor kappa-B). This extract also increased the expression of the antiapoptotic factor BCL2 (B-cell lymphoma 2) in the uterine tissue of rats administered BPA while impeding the abnormal expression of the tumor proteins p53, cylcin-D1, and BAX (BCL2-associated protein X). An enhanced steroidogenic event was supported by improved gonadotropins and reproductive hormone levels, feeble signaling of androgen receptors, and improved ovarian follicular growth with a distinct appearance of granulosa layer as well as better uterine histomorphology. The abundance of apigenin and catechin compounds in WG-ME may potentiate the above effects. The molecular interaction study predicted that apigenin inhibits TNF-α by interacting with its major site. Hence, WG-ME may exert its preventive efficacy in managing the functional imbalance of reproductive organs caused by BPA.Supplementary InformationThe online version contains supplementary material available at 10.1007/s13205-024-04117-0.

Odontogenic and anti-inflammatory effects of magnesium-doped bioactive glass in vital pulp therapy

AbstractThis study aimed to investigate the effects of magnesium-doped bioactive glass (Mg-BG) on the mineralization, odontogenesis, and anti-inflammatory abilities of human dental pulp stem cells (hDPSCs). Mg-BG powders with different Mg concentrations were successfully synthesized via the sol-gel method and evaluated using x-ray diffraction, Fourier-transform infrared spectroscopy, scanning electron microscopy, and transmission electron microscopy. Apatite formation was observed on the surfaces of the materials after soaking in simulated body fluid. hDPSCs were cultured with Mg-BG powder extracts in vitro, and no evident cytotoxicity was observed. Mg-BG induced alkaline phosphatase (ALP) expression and mineralization of hDPSCs and upregulated the expression of odontogenic genes, including those encoding dentin sialophosphoprotein, dentin matrix protein 1, ALP, osteocalcin, and runt-related transcription factor 2. Moreover, Mg-BG substantially suppressed the secretion of inflammatory cytokines (interleukin [IL]-4, IL-6, IL-8, and tumor necrosis factor-alpha). Collectively, the results of this study suggest that Mg-BG has excellent in vitro bioactivity and is a potential material for vital pulp therapy of inflamed pulps.

A multicenter evaluation of the QIAstat-Dx meningitis-encephalitis syndromic test kit as compared to the conventional diagnostic microbiology workflow

AbstractPurposeRapid diagnosis and treatment of infectious meningitis and encephalitis (ME) is critical to minimize morbidity and mortality. Recently, Qiagen introduced the CE-IVD QIAstat-Dx ME panel (QS-ME) for syndromic diagnostic testing of meningitis and encephalitis. Some data on the performance of the QS-ME in comparison to the BioFire FilmArray ME panel are available. In this study, the performance of the QS-ME is compared to the current diagnostic workflow in two academic medical centers in the Netherlands.MethodsA total of 110 cerebrospinal fluid samples were retrospectively tested with the QS-ME. The results obtained were compared to the results of laboratory-developed real-time PCR assays (LDTs), IS-pro, bacterial culture, and cryptococcal antigen (CrAg) testing. In addition, the accuracy of the QS-ME was also investigated using an external quality assessment (EQA) panel consisting of ten samples.ResultsFour of the 110 samples tested failed to produce a valid QS-ME result. In the remaining 106 samples, the QS-ME detected 53/53 viral targets, 38/40 bacterial targets, and 7/13 Cryptococcus neoformans targets. The discrepant bacterial results consisted of two samples that were previously tested positive for Listeria monocytogenes (CT 35.8) and Streptococcus pneumoniae (CT 40), respectively. The QS-ME detected one additional result, consisting of a varicella-zoster virus signal (CT 35.9), in a sample in which both techniques detected Streptococcus pyogenes. Finally, 100% concordance was achieved in testing a blinded bacterial ME EQA panel.ConclusionThe QS-ME is a relevant addition to the syndromic testing landscape to assist in diagnosing infectious ME.
