To compare the efficacy and safety of 100 mg CAI capsules in combination with vinorelbine + cisplatin and placebo in combination with vinorelbine + cisplatin as first-line treatment in patients with stage IV non-small cell lung cancer (NSCLC). Primary efficacy indicator: progression-free survival (PFS). Secondary efficacy indicators: objective response rate (CR+PR), disease control rate (CR+PR+SD), overall survival (OS), and quality of life.

Phase II Clinical and Pharmacologic Study of Radiation Therapy and CAI (Carboxy-Amido Triazole) in Adults With Newly Diagnosed Glioblastoma Multiforme

Drugs used in chemotherapy use different ways to stop tumor cells from dividing so they stop growing or die. Radiation therapy uses high-energy x-rays to damage tumor cells. Combining radiation therapy with chemotherapy may kill more tumor cells. Phase II trial to study the effectiveness of carboxyamidotriazole plus radiation therapy in treating patients who have newly diagnosed supratentorial glioblastoma multiforme.

High-Density Surface Electromyography Feedback Enhances Fibularis Longus Recruitment in Chronic Ankle Instability

ABSTRACTPurposeThis study aimed to determine whether individuals with chronic ankle instability (CAI) can activate the fibularis longus compartments with high-density surface electromyography (HD-sEMG) biofeedback to the same extent as those without CAI, and to analyze the effect of ankle position on compartment activation in individuals with CAI using HD-sEMG feedback.MethodsThere were 16 volunteers per group (CAI and No-CAI). The sEMG amplitude at each compartment (anterior and posterior) and the barycenter of the spatial sEMG amplitude distribution of the fibularis longus were recorded during eversion in neutral and plantar flexion positions at 30% and 70% of maximum voluntary contraction force, both with and without visual feedback on the spatial sEMG amplitude distribution.ResultssEMG amplitude of the posterior compartment of the fibularis longus in the CAI group trained with HD-sEMG feedback during eversion at 70% maximum voluntary contraction (in plantar flexion) was significantly higher than without HD-sEMG feedback (95% CI = 3.75–34.50% root mean square) and was similar to the activation of the No-CAI group (95% CI = −14.34% to 34.20% root mean square). Furthermore, individuals with CAI who underwent training with HD-sEMG feedback in plantar flexion exhibited a posterior displacement of the barycenter (95% CI = 0.56–2.84 mm).ConclusionsUtilizing HD-sEMG feedback during eversion in plantar flexion position increases activation of the fibularis longus posterior compartment in individuals with CAI to the same extent as healthy people. HD-sEMG–based topographic maps can serve as effective feedback training to restore motor control of the ankle. Long-term efficacy for improving motor function requires investigation through longitudinal studies.

Increased Femoral Neck Anteversion is Prevalent in Male Elite Youth Soccer Players with Chronic Ankle Instability

PurposeThe aim of this study was to compare femoral neck anteversion (FNA) and determine the prevalence of increased FNA in male elite youth soccer players with and without chronic ankle instability (CAI). Secondary aims were to evaluate the utility of FNA in predicting CAI and compare ankle and hip muscle strength in the two groups.Materials and methodsThe study included a total of 44 male elite youth soccer players, 22 with CAI (mean age 16.09 ± 1.34) and 22 without CAI (mean age 16.73 ± 1.28). FNA was measured with Craig's test, range of motion (ROM) was measured with a universal goniometer, and ankle and hip maximum voluntary isometric strength (MVIS) was measured with a handheld dynamometer.ResultsThe mean FNA angles of the CAI and control groups were 15.82° ± 1.44° and 12.09° ± 2.37°, respectively (p > 0.05). FNA was greater than 15° in 72% of the CAI group versus 4% of the control group (p < 0.05). A 1° increase in FNA was associated with threefold higher odds of having CAI (odds ratio 3.06, 95% confidence ratio: 1.37-6.81, p < 0.01). Mean ankle eversion and hip abduction MVIS values were 2.67 ± 0.52 Nm/kg and 3.83 ± 0.48 Nm/kg in the CAI group, compared to 3.03 ± 0.58 Nm/kg and 4.46 ± 0.98 Nm/kg in the control group, respectively (p < 0.05).ConclusionMale elite youth soccer players with CAI had greater FNA and were more likely to have increased FNA than those without CAI. They also exhibited ankle eversion and hip abduction muscle strength deficiencies compared to peers without CAI. FNA may be useful as a predictor of CAI in male elite youth soccer players.

Carbonic anhydrase inhibitors improves cognitive functions in Tg2576 AD mice

AbstractBackgroundOver the years, Alzheimer’s Disease (AD) has been identified as a multifactorial disease, with cerebral vascular dysfunction being one of the most common and early pathological features. Vascular risk factors (VRF) are thought to further increase AD risk and pathology. Cerebral Amyloid Angiopathy (CAA) is defined as the accumulation of amyloid‐beta (Aβ) on the vascular wall. CAA occurs progressively with age in human AD brains, vascular dementias, and the Tg2576 AD mouse model. The Tg2576 develops significant amounts of parenchymal Aβ plaques and vascular amyloid deposits making it a valuable model to study CAA and AD. To study the contribution of VRF to CAA and AD pathology, we induced hyperhomocysteinemia (HHcy) and hypertension (HP) in Tg2576 mice. We have shown that carbonic anhydrase inhibitors (CAi) decreased vascular amyloid accumulation and neuroinflammation and prevented cognitive impairment in TgSwDI mice. Here, we evaluated the effects of HP and HHcy in AD and CAA pathology. In addition, we assessed the beneficial effects of CAi in the Tg2576 with/without the presence of VRF.MethodsTo induce VRF, male and female WT and Tg2576 mice were fed a HHcy inducing diet and/or were given L‐NAME (NO inhibitor to induce HP) in the water starting at 5 months of age. To test the effects of CAi, the FDA‐approved CAi (Acetazolamide) was added to the diet starting at 5 months of age. Animals were then tested in a battery of behavioral measures including spatial learning and memory (barnes maze, fear conditioning, novel object recognition, open field, and rotarod), at 12 months of age.ResultThe presence of HHcy or HT worsens spatial learning and memory in the WT and Tg2576 mice. Preliminary results may suggest that treatment with CAi improved spatial learning and memory in the WT and Tg2576 mice, in the presence or absence of VRFs.ConclusionTaken together, our results suggest that VRF affect learning and memory in AD/CAA and non‐AD/CAA animal models. Importantly, we demonstrate that CAi is a promising treatment for treating mixed AD and CAA pathology.
