Treatment of COVID-19 Associated Olfactory Dysfunction: A Systematic Review

PURPOSE OF REVIEW:

COVID -19 associated olfactory dysfunction is widespread, yet effective treatment strategies remain unclear. This article aims to provide a comprehensive systematic review of therapeutic approaches and offers evidence-based recommendations for their clinical application.

RECENT FINDINGS:

A living Cochrane review, with rigorous inclusion criteria, has so far included 2 studies with a low certainty of evidence. In this systematic review we list clinical data of 36 randomised controlled trials (RCTs) and non-randomised studies published between Jan 1, 2020 and Nov 19, 2023 regarding treatment options for COVID-19 associated olfactory dysfunction. Nine treatment groups were analysed, including olfactory training, local and systemic corticosteroids, platelet-rich plasma (PRP), calcium chelators, vitamin supplements including palmitoylethanolamide with luteolin, insulin, gabapentin and cerebrolysin. Primary objective was the effect of the studied treatments on the delta olfactory function score (OFS) for objective/psychophysical testing. Treatments such as PRP and calcium chelators demonstrated significant improvements on OFS, whereas olfactory training and corticosteroids did not show notable efficacy for COVID-19 associated olfactory dysfunction.

Simultaneous analysis of four endocannabinoids and endocannabinoid congeners together with delta-9 tetrahydrocannabinol and related metabolites in dried blood spots

In the medical field, the endocannabinoid system is both a difficult and hot topic for scientific and media reasons (cannabis, cannabidiol, etc.). Due to their instability and low concentrations in blood and physicochemical properties, the complexity of endocannabinoid assay is previously reported in different matrices. This study aims to report a LC-MS/MS method for the simultaneous quantification of four endocannabinoids and endocannabinoid congeners: N-arachidonoylethanolamide (anandamide or AEA), 2-arachidonoylglycerol/1-arachidonoylglycerol (2-AG/1-AG), oleoylethanolamide (OEA) and palmitoylethanolamide (PEA), together with Δ9-tetrahydrocannabinol (THC) and its main metabolites [11-nor-9-carboxy-Δ9-tetrahydrocannabinol (THC-COOH) and 11-hydroxy- Δ9-tetrahydrocannabinol (11-OH THC] in Dried Blood Spot (DBS). This validated method reported LLOQs of 0.1 µg/L for AEA, 0.5 µg/L for THC and 11-OH THC, and 1 µg/L for OEA, PEA, 2AG and THC-COOH. The method demonstrated intra- and interassay accuracy and precision of less than 20 % for the LLOQ, and less than 15 % for the other calibration points. In particular, the stability carried out in this study demonstrated the practical applicability of DBS for the measurement of endocannabinoids in the whole blood. Therefore, we propose this method as a tool for basic and clinical research to support the growing interest of the scientific community in the endocannabinoid system.

As a significant reactive oxygen species (ROS), ClO^- plays versatile roles in daily life and many biological events. However, its abnormal levels are responsible for serious harm to human health. Therefore, it is of crucial interest to develop effective methods for detecting ClO^- in foods and living organisms. In this work, a novel fluorescent probe ethyl 2-(3-formyl-2-methoxy-10H-phenothiazin-10-yl)acetate (PEA) for detecting ClO^- was presented. It shows the merits of rapid ratiometric response (within 10 s), high selectivity and very large Stokes shift (190 nm). The detection limit of PEA for ClO^- was determined to be 0.47 μM. We not only successfully prepared paper test strips for efficient qualitative naked eye ClO^- detection, but also demonstrated its potential for the valid detection of ClO^- in natural water samples, beverages and foods. Furthermore, the probe PEA was also applied to the fluorescence imaging of ClO^- in onion epidermal cells.

Quantum BioPharma Receives Ethics Committee Approval for Phase 2 Clinical Trial of FSD202 for Nociplastic Pain in Patients with Idiopathic Mast Cell Activation Syndrome

TORONTO, May 28, 2025 (GLOBE NEWSWIRE) -- Quantum BioPharma Ltd. MCAS refers to a group of disorders characterized by multisystem symptoms resulting from the accumulation of altered mast cells and/or abnormal mast cell mediator release, causing repeated anaphylactic symptoms/episodes and trapping a patient in a cycle of neurogenic pain and inflammation. Chronic widespread musculoskeletal nociplastic pain, a type of chronic pain characterized by altered pain perception that is not directly due to tissue injury, is associated with MCAS. The cause of MCAS is unknown and there currently is no cure. FSD202, a drug product containing ultra-micronized palmitoylethanolamide (PEA), has the potential to address a wide range of inflammatory diseases in addition to MCAS.
