The purpose of the research is to determine the content of berberine hydrochloride and paeoniflorin in Qingweizhitong pellet by high performance liquid chromatog. in different conditions.The determination for berberine hydrochloride was conducted by Diamonsil-C18 column(250 mm*4.6 mm, 5μm), the mobile phase was acetonitrile-0.033 mol·L-1 potassium dihydrogen phosphate(30:70 for volume), the detection wavelength was 345 nm.The determination for paeoniflorin was conducted by Shimpack C18(150 mm*4.6 mm, 5 μm), the mobile phase was acetonitrile -0.3% phosphoric acid(12.7:87.3 for volume), the detection wavelength was 230 nm.Good linear relationship between area and amount was noted in the range of 0.016-0.080 μg for berberine hydrochloride with a correlation coefficient of 0.9998.The average recovery of berberine hydrochloride was 98.68%, and the RSD was 0.74%.Good linear relationship between area and amount was noted for 0.268-1.340 μg for paeoniflorin with a correlation coefficient of 0.9999.The average recovery of paeoniflorin was 99.03%, and the RSD was 0.79%.The established method is simple, accurate, sensitive and good repeatability, which can be applied for the quality control of Qingweizhitong pellet.
