A Prospective, Non-interventional, Single-Centre Study Assessing the Time Course of a Holistic Perioral Rejuvenation Procedure

The goal of this observational study is to learn how a perioral rejuvenation procedure affects perioral line severity and lip volume over time in adults undergoing routine aesthetic treatment in a single clinical center. The main questions it aims to answer are:
* What proportion of participants achieves a ≥1-point improvement from baseline on both the Lip Fullness Scale (LFS) and the Static Perioral Lines Assessment Scale (SPLAS) at Week 12?
* How do skin wrinkling/surface roughness, lip and perioral volume, and patient satisfaction change over 6 and 12 weeks?
Participants will:
* Receive a routine (non-study-mandated) holistic perioral rejuvenation procedure as decided by their treating clinician.
* Attend study visits at approximately baseline, Week 6, and Week 12.
* Undergo independent clinical assessments (assessor not involved in treatment delivery) of LFS and SPLAS.
* Have skin wrinkling and surface roughness documented by VISIA skin analysis (baseline, Week 6, Week 12).
* Have lip and perioral volume measured by VECTRA M3 volume analysis (baseline, Week 6, Week 12).
* Complete a standardized patient satisfaction questionnaire (Week 12).

A Prospective, Non-interventional, Single-Centre Study Assessing the Time Course of a Holistic Extended Periorbital Rejuvenation Procedure

The goal of this observation is to assess the time course of holistic extended periorbital rejuvenation using botulinum toxin A, cross-linked hyaluronic acid and polynucleotides corresponding to the application in routine practice in adults. It will also document the safety of treatment procedure and patient satisfaction.
The main questions it aims to answer are:
* How does a holistic extended periorbital rejuvenation procedure using botulinum toxin A, cross-linked hyaluronic acid filler, and polynucleotides change skin wrinkling around the eye area over time (particularly after 8 and 22 weeks)?
* How does the treatment affect the severity of dynamic glabellar lines at 8 and 22 weeks?
* Does the procedure improve midface volume deficiency after 8 and 22 weeks?
* How satisfied are participants with the overall rejuvenation treatment?
The investigator will observe the combination treatment of a drug Letybo® and medical devices PhilArt® Eye and Saypha® Volume Plus Lidocaine within their approved indications (non-interventional study, on-label use). The combination treatment aims to rejuvenate and improve the eye area holistically in patients coming routinely to the clinic.
Participants will:
* take PhilArt® Eye on Week 0 (Baseline), 3 and 6; Saypha® Volume Plus Lidocaine on Week 0 (Baseline); Letybo® on Week 6.
* visit the clinic on Week 0 (Baseline), 3, 6, 8 and 22 as per clinical practice for checkups and satisfaction assessment.

Diffusion Characteristics of LetibotulinumtoxinA, OnabotulinumtoxinA, and AbobotulinumtoxinA and its Impact on Muscle Relaxation: A Randomized Split-Face Clinical Trial

INTRODUCTION:

Injections with botulinumtoxinA (BoNTA) are the most common nonsurgical aesthetic procedure but carry risks such as unintended muscle diffusion. Understanding formulation behavior is key to optimizing safety and efficacy. This study compares the diffusion characteristics from letibotulinumtoxinA (Leti-BoNTA) to onabotulinumtoxinA (Ona-BoNTA) and abobotulinumtoxinA (Abo-BoNTA) and its impact on muscle relaxation of frontalis muscle.

METHODS:

In a double-blind, randomized trial, 30 healthy adults received Leti-BoNTA on one forehead side and either Ona-BoNTA or Abo-BoNTA on the other. Anhidrosis was measured via Minor's starch test over 6 months, and wrinkle severity was assessed using standardized photography and the Croma Scale.

RESULTS:

Leti-BoNTA had a significantly smaller maximal anhidrotic area than Ona-BoNTA (-15.1 ± 5.5 cm², p < 0.001) and Abo-BoNTA (-25.2 ± 14.5 cm², p < 0.001). An area under the curve (AUC) analysis confirmed the largest area of anhidrosis for Abo-BONTA over the 6-month period, followed by Ona-BONTA and Leti-BoNTA. Despite its lower diffusion, Leti-BoNTA effectively improved wrinkles, with ≥ 1-point improvement in 92% of subjects by week 2.

CONCLUSIONS:

Leti-BoNTA provides wrinkle reduction with a more limited diffusion, as assessed with the Minor's starch-iodine test. Its precise action in this trial implies to be a viable option when controlled spread is essential.

LetibotulinumtoxinA Attenuates the Psychological Burden of Glabellar Lines and Is Associated With High Subject Satisfaction in Phase 3 Clinical Trials

BACKGROUND:

Hyperfunctional glabellar frown lines can transmit facial miscues that adversely affect emotional communication, increase perceptions of age, and diminish self-esteem.

OBJECTIVE:

To evaluate the efficacy of letibotulinumtoxinA in mitigating the negative psychological impact associated with moderate to severe glabellar lines and to assess subject satisfaction with treatment outcome in the BLESS phase 3 clinical trials.

MATERIALS AND METHODS:

Baseline and posttreatment assessments were made using validated subject-administered instruments: Modified Skindex-16 Glabellar Line Quality of Life (GL-QoL) Scale, Facial Assessment and Cosmetic Evaluation Questionnaire (FACE-Q) Appraisal of Lines Between Eyebrows Scale, FACE-Q Age Appraisal Visual Analog Scale, and FACE-Q Satisfaction with Outcome Scale. An integrated analysis using pooled BLESS data was conducted on these secondary end points.

RESULTS:

Among enrolled and treated subjects (N = 1,272), 85.5% had moderate to severe psychological impact at baseline. LetibotulinumtoxinA subjects experienced significant improvements compared with placebo on all measures. Mean improvement to Week 4 for the Modified Skindex-16 GL-QoL Scale overall score was −33.84 for letibotulinumtoxinA subjects compared with −1.37 for placebo subjects (p < .001). Attenuation of psychological burden was highly correlated with improvement in glabellar line severity (p < .0001).

CONCLUSION:

LetibotulinumtoxinA significantly improved the psychosocial burden associated with glabellar lines across all trials. Treated subjects experienced improved quality of life, younger perceived age, and satisfaction with treatment outcome.

Abstract:

We trained a deep learning algorithm to use skin optical coherence tomography (OCT) angiograms to differentiate between healthy and type 2 diabetic mice. OCT angiograms were acquired with a custom‐built OCT system based on an akinetic swept laser at 1322 nm with a lateral resolution of ∼13 μm and using split‐spectrum amplitude decorrelation. Our data set consisted of 24 stitched angiograms of the full ear, with a size of approximately 8.2 × 8.2 mm, evenly distributed between healthy and diabetic mice. The deep learning classification algorithm uses the ResNet v2 convolutional neural network architecture and was trained on small patches extracted from the full ear angiograms. For individual patches, we obtained a cross‐validated accuracy of 0.925 and an area under the receiver operating characteristic curve (ROC AUC) of 0.974. Averaging over multiple patches extracted from each ear resulted in the correct classification of all 24 ears.
