[Translation]

This study aimed to investigate the pharmacokinetic characteristics of netupitant palonosetron capsules (0.3 g/0.5 mg) developed and produced by Zhejiang Kanglaite Pharmaceutical Co., Ltd. after single oral administration on an empty stomach or after a meal. Using netupitant palonosetron capsules (Okangze®, 0.3 g/0.5 mg) produced by Helsinn Birex Pharmaceuticals Ltd. as the reference preparation, the pharmacokinetic parameters Cmax and AUC0-72 of the two preparations were compared, and the bioequivalence of the two preparations in humans was evaluated.

A Simple and Accurate Method for the Determination of Related Substances in Coenzyme Q10 Soft Capsules

As a new dosage form, coenzyme Q10 (Co-Q10) soft capsules are easily absorbed and utilized by the human body. Co-Q10 soft capsules can effectively improve the bioavailability and reduce medical costs for patients. A main concern about Co-Q10 as an active pharmaceutical ingredient (API) is how to control the total quantity of related substances. In this article, according to the degradation pattern of the API, the most easily degradable impurity (impurity X) in the sample was prepared and its chemical structure was determined. Furthermore, a simple and accurate method was developed for the determination of related substances and to avert the interference of excipient ingredients in Co-Q10 soft capsules. The approach was validated adequately and the primary impurity X was confirmed accurately. The limit of total quantity of related substances (less than 1%) could be revised to the level of specific impurity X being no more than 0.5%, in this effective quality control method of Co-Q10 soft capsules. The revised level is suggested to be included in the corresponding standard of the supplement taken from the Pharmacopoeia of the People’s Republic of China (2015 edition). This can provide a feasible method for the relevant enterprises and regulatory authorities to control the related substances of coenzyme Q10 soft capsules.

Determination of spinosin and 6'''-feruloylspinosin in rat plasma after oral administration of flavonoid extract from Ziziphi Spinosae Semen by SPE-HPLC-DAD

A convenient and sensitive method for determination of flavonoid metabolites in rat plasma after oral administration of Ziziphi Spinosae Semen extract was established.The rat blood samples were collected from each rat by puncture of the retro-orbital sinus from 0 to 24 h after oral administration of flavonoid extract of jujube seeds, then pretreated with a Waters OASIS TM SPE column using cartridges.The plasma samples were analyzed by high-performance liquid chromatog. with diode array detector (HPLC-DAD) method to detect the main metabolites.The original flavonoids were the major forms of flavonoid metabolites in rat plasma.The calibration curves were linear over the range of 0.52 119.3 μg·mL^-1 (r > 0.9998) for spinosin and 0.38-91.7 μg·mL^-1 (r > 0.9997) for 6'''-feruloylspinosin with the mean extraction recoveries as 84.76% and 84.33%, resp.The average plasma drug concentration of them reached the top level at the 12th and 24th hour after oral administration, and disappeared gradually in vivo.This convenient solid-phase extraction (SPE)-HPLC-DAD method could be used to determinate the flavonoid metabolites of Ziziphi Spinosae Semen.The pharmacokinetic feature of main active components of traditional Chinese medicine in vivo could be characterized as well.
