Introduction

Catalyzes the GTP-dependent ribosomal translocation step during translation elongation (PubMed:26593721). During this step, the ribosome changes from the pre-translocational (PRE) to the post-translocational (POST) state as the newly formed A-site-bound peptidyl-tRNA and P-site-bound deacylated tRNA move to the P and E sites, respectively (PubMed:26593721). Catalyzes the coordinated movement of the two tRNA molecules, the mRNA and conformational changes in the ribosome (PubMed:26593721).

Comparing the malignant properties of parental and a knock-in version of HCT116 cell line expressing the CDK2-mutant of eukaryotic elongation factor 2 (eEF2)

BACKGROUNDModulation of protein synthesis according to the physiological cues is maintained through tight control of Eukaryotic Elongation Factor 2 (eEF2), whose unique translocase activity is essential for cell viability. Phosphorylation of eEF2 at its Thr56 residue inactivates this function in translation. In our previous study we reported a novel mode of post-translational modification that promotes higher efficiency in T56 phosphorylation. Cyclin A/CDK2-mediated phosphorylation of eEF2 at the S595 residue is required for more potent phosphorylation at the Thr56, suggesting CDK2 takes a role in robust suppression of protein synthesis.METHODS AND RESULTSIn the current study, we analyzed the cell cycle, proliferation, cell death, migration, colony formation, autophagy, and response to Cisplatin properties of the point-mutant variant of HCT116 cells that express the CDK2 mutant (S595A-eEF2) of eEF2. The knocked in S595A mutation resulted in decreased levels of T56 phosphorylation of eEF2, which appears to have similar biological consequences to other experimental manipulations such as silencing the activity of the kinase for the Thr56 residue, eEF2 Kinase (eEF2K).CONCLUSIONOur findings indicate that interfering with the inhibition of eEF2 results in elevated protein synthesis in HCT116 cells and is associated with the progression of malignancy in the colorectal cancer cell line, where eEF2K activity could provide a tumor suppressive role.

Antibiotics/coccidiostat exposure induces gut-brain axis remodeling for Akt/mTOR activation and BDNF-mediated neuroprotection in APEC-infected turkeys

The poultry industry relies extensively on antibiotics and coccidiostats as essential tools for disease management and productivity enhancement. However, increasing concerns about antimicrobial resistance (AMR) and the toxicological safety of these substances have prompted a deeper examination of their broader impacts on animal and human health. This study investigates the toxicological effects of antibiotics and coccidiostats on the gut-brain axis and microbiota in turkeys, with a particular focus on molecular mechanisms that may influence neurochemical and inflammatory responses. Our findings reveal that enrofloxacin exposure leads to the upregulation of BDNF, suggesting a neuroprotective effect, while monensin treatment significantly increased eEF2 kinase expression, indicative enhanced neuronal activity. In turkeys infected with Avian Pathogenic Escherichia coli (APEC), early administration of doxycycline and monensin significantly upregulated the mTOR/BDNF and Akt/mTOR pathways, along with elevated histamine levels, underscoring their role in inflammatory responses modulation. However, treatments administered at 50 days post-hatch did not significantly alter protein levels, though both enrofloxacin and monensin increased serotonin and dopamine levels, suggesting potential neurotoxicological impacts on mood and cognitive functions. These results highlight the complex interactions between antibiotic use, gut microbiota alterations, and neurochemical pathways, with toxicological implications for environmental pollution and public health. This research provides critical insights into the potential toxic effects of prolonged antibiotic and coccidiostat exposure in poultry production, emphasizing the need for responsible use to mitigate risks to ecosystems and human health.

Macrophage Dvl2 deficiency promotes NOD1-Driven pyroptosis and exacerbates inflammatory liver injury

Dishevelled 2 (Dvl2) is a key mediator of the Wingless/Wnt signaling pathway that regulates cell proliferation, migration, and immune function. However, little is known about the role of macrophage Dvl2 in modulating NOD1-mediated pyroptosis and hepatocyte death in oxidative stress-induced inflammatory liver injury. In a mouse model of oxidative stress-induced liver inflammation, mice with myeloid-specific Dvl2 knockout (Dvl2^M-KO) displayed exacerbated ischemia/reperfusion (IR) stress-induced hepatocellular damage with increased serum ALT levels, oxidative stress, and proinflammatory mediators. Unlike in Dvl2^FL/FL controls, Dvl2^M-KO enhanced NOD1, caspase-1, GSDMD, and NF-κB activation in liver macrophages after IR. Interestingly, IR stress enhanced YAP colocalized with HSF1 in Dvl2^FL/FL macrophages, while macrophage Dvl2 deficiency reduced YAP and HSF1 colocalization in the nucleus under inflammatory conditions. Importantly, Dvl2 deletion diminished nuclear YAP interacted with HSF1 and augmented NOD1/caspase-1 and GSDMD activation in response to inflammatory stimulation. However, Dvl2 activation increased YAP interaction with HSF1 and activated HSF1 target gene eEF2, inhibiting NOD1/caspase-1, GSDMD, and NF-κB activity. Moreover, macrophage eEF2 deletion increased the NOD1-caspase-1 interaction, GSDMD activation, HMGB1 release, and hepatocyte LDH release after macrophage/hepatocyte co-culture. Adoptive transfer of eEF2-expressing macrophages in Dvl2^M-KO mice alleviated IR-triggered liver inflammation and hepatocellular damage. Therefore, macrophage Dvl2 deficiency promotes NOD1-mediated pyroptosis and exacerbates IR-induced hepatocellular death by disrupting the YAP-HSF1 axis. eEF2 is crucial for modulating NOD1-driven pyroptosis, inflammatory response, and hepatocyte death. Our findings underscore a novel role of macrophage Dvl2 in modulating liver inflammatory injury and imply the therapeutic potential in organ IRI and transplant recipients.
