Introduction

This is the catalytic component of the active enzyme, which catalyzes the hydrolysis of ATP coupled with the exchange of sodium and potassium ions across the plasma membrane. This action creates the electrochemical gradient of sodium and potassium ions, providing the energy for active transport of various nutrients (PubMed:29499166, PubMed:30388404). Could also be part of an osmosensory signaling pathway that senses body-fluid sodium levels and controls salt intake behavior as well as voluntary water intake to regulate sodium homeostasis (By similarity).

An in silico analysis of dicofol-induced neurotoxicity mechanisms in humans

Dicofol (DCF) is an organochlorine pesticide that has recently been recognized as a persistent organic pollutant. This study begins by investigating the neurotoxicity of DCF and its metabolites through in silico tools. It subsequently explores the molecular mechanisms and key targets associated with DCF-induced neurotoxicity in humans by employing network toxicology, multi-level bioinformatics approaches, and molecular docking analyses. The prediction results indicate that both DCF and its metabolites can traverse the blood-brain barrier, penetrating the central nervous system, and inducing neurotoxicity. A thorough analysis has identified 56 potential targets linked to DCF-induced neurotoxicity. Gene Ontology enrichment analysis revealed significant associations with pathways related to sodium ion transmembrane transport, sodium/potassium-exchanging ATPase complexes, and P-type calcium transporter activity. Pathway enrichment analysis suggests that DCF-induced neurotoxicity arises from disruptions in ion transport via P-type ATPases. Further examination of gene-gene and protein-protein interactions, along with centrality analysis, identified 11 hub targets, including ATP1A1, ATP1A2, ATP1A3, ATP1A4, ATP1B1, ATP1B2, and MAPK1, as key players. Notably, six of these targets are subunits of the Na⁺/K⁺-ATPase, a P-type ATPase. Molecular docking results demonstrated that DCF binds more effectively to the ATP1A3-ATP1B1 protein complex than to its natural ligand, ATP. These findings suggest that DCF may inhibit Na⁺/K⁺-ATPase through ATP1A3, resulting in an imbalance of sodium and potassium gradients and ultimately leading to neurotoxicity.

Components in Prunus mume and their targets in the alleviation of severe depression identified using network pharmacology analysis

BACKGROUNDPrunus mume (PM), a traditional Chinese medicinal herb, is well known for relieving depression.STUDY DESIGNTo elucidate its potential therapeutic benefits for major depressive disorder (MDD), further research is required to investigate its underlying mechanisms.MATERIALS AND METHODSUsing UPLC-QE-MS and the Traditional Chinese Medicine Systems Pharmacology Database (TCMSP), we analyzed PM's ingredients and targets. The Gene Expression Omnibus (GEO) database identified MDD-related targets. Enrichment analysis identified key components for MDD treatment, refined via protein-protein interaction (PPI) networks and machine learning. Molecular docking with AutoDock software assessed binding affinity.RESULTSWe identified 35 PM components and 691 targets. Bioinformatics revealed 1,105 differentially expressed genes in MDD. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses showed significant enrichment in protease binding, protein tyrosine kinase activity, and pathways like mTOR, Ras, and VEGF. PPI networks and machine learning identified five hub genes - SELP, MMP8, HK2, ATP1A1, and DAO - as therapeutic targets. Molecular docking analysis showed that PM ingredients - Quercetin, Luteolin, Tamarixetin, Isorhamnetin, 8-Hydroxykaempferol, Morin - bind effectively to key targets.CONCLUSIONThis study elucidates active components and potential mechanisms for treating MDD with PM, providing insights for future therapeutic targets and drug development.

Target Identification of Marine Natural Product Odoamide:Odoamide Induces Apoptotic Cell Death by Targeting ATPase Na⁺/K⁺ Transporting Subunit Alpha 1 (ATP1A1)

AbstractMarine natural products show a large variety of unique chemical structures and potent biological activities. Elucidating the target molecule and the mechanism of action is an essential and challenging step in drug development starting with a natural product. Odoamide, a member of aurilide‐family isolated from Okinawan marine cyanobacterium, has been known to exhibit highly potent cytotoxicity against various cancer cell lines. In this study, we investigated the target protein and the cytotoxic mechanism of odoamide. Compared to existing anticancer agents, odoamide showed a unique fingerprint in the JFCR39 cancer cell panel and a characteristic pattern in gene expression profiling. Affinity chromatography utilizing a biologically active odoamide probe identified ATPase Na+/K+ transporting subunit alpha 1 (ATP1A1) as a specific binding protein. Additionally, cells resistant to odoamide were found to have mutations at Gly98 and Gly99 of the ATP1A1 protein. The apparently attenuated cytotoxic and apoptotic activities of odoamide in odoamide‐resistant cells suggests that the induction of these activities by odoamide is critically dependent on its interaction with ATP1A1. We conclude that odoamide induces apoptotic cell death by targeting ATP1A1, and we discuss the impact of affinity‐based target identification for natural products and the potential of ATP1A1 inhibitors for drug discovery.
