Introduction

Cleaves C-terminal arginine or lysine residues from biologically active peptides such as kinins or anaphylatoxins in the circulation thereby regulating their activities. Down-regulates fibrinolysis by removing C-terminal lysine residues from fibrin that has already been partially degraded by plasmin.

A Phase I, Single Blind, Placebo-controlled, Randomized Study to Assess the Safety of DS-1040b in Subjects With Thrombectomy Treated Acute Ischemic Stroke.

The aim of this study is to find out if DS-1040b is safe and tolerable in acute ischemic stroke patients with thrombectomy. Four groups will receive different doses of DS-1040b by intravenous infusion for 6 hours. Groups with the lowest dose will start. When it is determined that each dose is safe and tolerable, the next higher dose will be given to the next group.

Genomic and antimicrobial resistance profiles of Clostridium perfringens isolated from pets in China

Clostridium perfringens is a notable pathogen causing diarrhea in domestic animals. However, data on this pathogen's prevalence and genomic characteristics in pets are limited. We collected 300 fecal samples from companion animals across two cities in China and isolated 150 strains for genomic sequencing and antimicrobial susceptibility testing (AST). Our findings showed a high prevalence of two key virulence genes (VGs), pfoA (77.33 %, 116/150) and cpb2 (60.67 %, 91/150). Moreover, for the first time in China, we identified a strain carrying netF. The analysis of AST and genomic antimicrobial resistance genes (ARGs) highlighted the resistance patterns of pet-derived C. perfringens strains in China. Particularly, we observed a high resistance rate to erythromycin, primarily associated with erm(Q), which was found in 88.67 % (133/150) of all isolates. This resistance rate was higher than that reported in previous studies. Genetic context analysis identified a novel plasmid group harboring the erm(Q) gene. In summary, this study revealed the antibiotic resistance and genomic characteristics of C. perfringens strains derived from pets in China, providing a reference for the prevention of related diseases and further research. Notably, these findings underscore the need for continuous monitoring of resistance trends, particularly concerning the spread of erm(Q), to lessen the impact of antimicrobial resistance in veterinary medicine.

Functional assessment of genetic variants in thrombomodulin detected in patients with bleeding and thrombosis

AbstractThrombomodulin (TM) expressed on endothelial cells regulates coagulation. Specific nonsense variants in the TM gene, THBD, result in high soluble TM levels causing rare bleeding disorders. In contrast, although THBD variants have been associated with venous thromboembolism, this association remains controversial. A multigene panel was used to diagnose 601 patients with inherited bleeding or thrombotic disorders. This resulted in the identification of 8 THBD variants for 6 patients with a thrombotic (C175S, A282P, L433P, P501L, G502R, and P508L) and 2 patients with a bleeding (P260A and T478I) phenotype. These were all classified as variants of uncertain significance, and we here aimed to assess their functional role in coagulation. For this purpose, soluble and cell membrane–bound recombinant TM were produced in Expi293F cells. L433P TM showed a marked decrease in the inhibition of thrombin generation and complete inhibition of protein C and thrombin activatable fibrinolysis inhibitor (TAFI) activation. Soluble C175S TM showed decreased inhibition of thrombin generation and protein C activation, whereas no effect was observed for cell membrane-bound recombinant TM. For the other TM variants, no effect on thrombin generation, protein C, or TAFI activation could be observed. Surface plasmon resonance analysis showed no thrombin-TM binding in the presence of L433P because this residue is located at their interaction site. In conclusion, our study shows the functional effects of L433P TM and potentially C175S TM, which are compatible with an increased thrombosis risk. THBD variants are rare but can be relevant to both bleeding and thrombosis. Functional assays for these variants are critical to understand their roles.

A novel strategy for rapid, portable, and sensitive detection of Clostridium perfringens cpb2 gene using smartphone-based electrochemical DNA biosensor based on screen-printed electrodes modified with nanocomposite

An accurate, sensitive, low-cost, portable, and easy-to-use method for the quantitative detection of Clostridium perfringens (Cp), a zoonotic pathogen widely found in nature and capable of spreading through contaminated food or environments, is essential for epidemiology, prevention, and diagnostics. Here, we have designed a smartphone-based electrochemical DNA biosensor, which utilizes carboxyl-functionalized multi-walled carbon nanotubes (COOH-MWCNTs) as the substrate, L-cysteine (L-Cys) as the linker, and a synergistic modification of the bioelectrode with gold nanoparticles (AuNPs) and polyamidoamine dendrimers (PAMAM). This design enables highly sensitive, low-cost, label-free, and portable detection of the cpb2 gene in Cp. The sensor not only enhances detection performance but also improves convenience and practicality. Differential pulse voltammetry (DPV) was used to monitor the electrochemical signal response to changes in target DNA concentration, enabling the tracking of the DNA hybridization process. Under optimized conditions, the biosensor exhibited a linear detection range from 10⁻¹⁴ to 10⁻⁸ M, with a detection limit of 1.5 fM. It demonstrated excellent selectivity for the cpb2 gene and was successfully applied to detect variations in cpb2 gene content in wastewater and fecal DNA samples. Compared with the traditional method, the detection time of this method is short, the operation of professional and technical personnel is not required, the instrument is small and portable, and the single detection cost is significantly reduced. This study provides a new strategy for the rapid, portable, and highly sensitive detection of bacterial toxin genes in livestock and aquaculture.
