Introduction

Component of the COP9 signalosome complex (CSN), a complex involved in various cellular and developmental processes. The CSN complex is an essential regulator of the ubiquitin (Ubl) conjugation pathway by mediating the deneddylation of the cullin subunits of SCF-type E3 ligase complexes, leading to decrease the Ubl ligase activity of SCF-type complexes such as SCF, CSA or DDB2. The complex is also involved in phosphorylation of p53/TP53, c-jun/JUN, IkappaBalpha/NFKBIA, ITPK1 and IRF8/ICSBP, possibly via its association with CK2 and PKD kinases. CSN-dependent phosphorylation of TP53 and JUN promotes and protects degradation by the Ubl system, respectively.

CsCIPK20 Improves Tea Plant Cold Tolerance by Modulating Ascorbic Acid Synthesis Through Attenuation of CsCSN5‐CsVTC1 Interaction

ABSTRACTLow temperature is a limiting environmental factor for tea plant growth and development. CBL‐interacting protein kinases (CIPKs) are important components of the calcium pathway and involved in plant development and stress responses. Herein, we report the function and regulatory mechanisms of a low‐temperature‐inducible gene, CsCIPK20, in tea plants. The overexpression of CsCIPK20 in Arabidopsis and its transient knockdown in tea plants confirmed its positive role in cold resistance. Notably, the ascorbic acid (AsA) levels increased in the overexpression lines and decreased in the CsCIPK20 knockdown tea plants under freezing stress. Transcriptomic analysis revealed that genes involved in flavonoid metabolism, glutathione metabolism, and AsA biosynthesis were significantly regulated by CsCIPK20. Furthermore, we found that CsCSN5, a key component of the COP9 signalosome, interacted with CsCIPK20 to mediate CsCIPK20 degradation. CsCSN5 interacted with CsVTC1, a key enzyme in AsA biosynthesis, and mediated CsVTC1 degradation. Knockdown of CsVTC1 in tea plants enhanced sensitivity to low temperature. Moreover, we demonstrated that CsCIPK20 competed with CsVTC1 to bind to CsCSN5, which protected CsVTC1 from degradation mediated by CsCSN5 and contributed to AsA accumulation. Overall, our findings uncovered a mechanistic framework through which the CsCIPK20‐CsCSN5‐CsVTC1 module mediated AsA accumulation and low‐temperature resistance in tea plants.

PKS1 involved in anthocyanin accumulation in red-skinned pear fruit

KEY MESSAGEPcPKS1 can prevent PcCSN5a from acting as an inhibitor of anthocyanin synthesis by binding to PcCSN5a, ultimately leading the accumulation of anthocyanins. Light is a crucial environmental factor that regulates anthocyanin accumulation in plants. However, the molecular mechanisms by which light signals influence anthocyanin accumulation in fruits have not yet been fully elucidated. We identified the differentially expressed gene Pyrus communis PHYTOCHROME KINASE SUBSTRATE 1 (PcPKS1), which is associated with anthocyanin accumulation in plants, in a previous study. Through measurements of the expression of PcPKS1 in 'Starkrimson' and 'Red Bartlett' pear fruit at various developmental stages and in different pear varieties, quantitative and transient expression experiments conducted on red and green skin tissues confirmed the relationship between PcPKS1 and anthocyanin accumulation. Pyrus communis COP9 SIGNALOSOME COMPLEX SUBUNIT 5A (PcCSN5a) protein, which interacts with PcPKS1, was identified from a yeast library screening. The interaction between the two proteins was validated through yeast two-hybrid (Y2H), bimolecular fluorescence complementation (BiFC), and split-luciferase (Split-LUC) experiments. Subcellular localization and co-localization experiments revealed that PcPKS1 was localized to the cell membrane, whereas PcCSN5a was localized to the cell membrane and nucleus, with PcPKS1 and PcCSN5a co-localized on the cell membrane. Transient expression in strawberry fruit indicated that PcPKS1 positively regulated anthocyanin accumulation, whereas PcCSN5a negatively regulated anthocyanin accumulation and diminished the capacity of PcPKS1 to promote anthocyanin accumulation. This study provides novel insights into the molecular mechanisms underlying light-regulated anthocyanin accumulation in red-skinned pear fruit.

Ubiquitin‐mediated degradation of the inhibitor FvMYB1 and the activator FvBBX20 by FvCSN5 balances anthocyanin biosynthesis in strawberry fruit

SUMMARYPlant CSN5 is widely recognized as the subunit of the COP9 signalosome and CSN5 is mainly involved in plant growth and development, and tolerance to biotic and abiotic stresses. However, the molecular mechanism of CSN5 regulating anthocyanin biosynthesis in plants is still largely unknown. Here, we identified FvCSN5 from the woodland strawberry yeast two‐hybrid library using the anthocyanin pathway inhibitor MYB1 as bait. We demonstrated the interaction of FvCSN5 and FvMYB1 by H2Y, Pull‐down, LCI, and BiFC assays. FvCSN5 was expressed in all test tissues and localized in the nucleus and cytosol with self‐activation activity. Stable overexpression of FvCSN5 in woodland strawberries reduced anthocyanin accumulation in fruits. The protein level of FvMYB1 greatly decreased in overexpressing FvCSN5 plants compared with wild‐type plants. Protein degradation assay and MG‐132 treatment (a proteasome inhibitor blocking 26S proteasome activity) revealed FvCSN5 degraded FvMYB1 through the ubiquitination pathway. In addition, FvCSN5 also interacted with the anthocyanin activator FvBBX20 and FvBBX20 could be degraded by FvCSN5. Moreover, transient expression analysis showed the expression of anthocyanin biosynthetic genes FvCHS and FvF3H was greatly increased and decreased when FvCSN5 was co‐expressed with FvMYB1 and FvBBX20, respectively. These results indicate that FvMYB1‐FvCSN5‐FvBBX20 is a novel ternary complex that regulates anthocyanin biosynthesis by the ubiquitination pathway.
