Acute exposure of perchlorate on zebrafish larvae: Neurotoxicity during development

OBJECTIVEThis study aimed to assess the acute developmental toxicity of perchlorate exposure and identify its manifestation of neurotoxicity in zebrafish (Danio rerio) model.METHODSZebrafish larvae were exposed to sodium perchlorate at early developmental stages and the phenotypes of various developmental abnormalities were observed and recorded. Neurodevelopmental toxicity has been studied in particular in terms of central nervous system apoptosis, peripheral motor abnormalities, and abnormal autonomic motor behavior. Samples were processed for pathological, transcriptomic, and PCR analyses.RESULTSAcute exposure of zebrafish larvae to perchlorate resulted in developmental toxicity in a concentration-dependent manner. Heart rate abnormality (33 %), slow blood flow (37 %), abnormal size of liver (23 %), delayed yolk absorption (70 %), and abnormal body pigmentation (100 %) were observed in the 3.83 mg/mL group. Heart edema (10 %), heart rate abnormality (60 %), slow blood flow (67 %), abnormal size of liver (77 %), delayed yolk absorption (100 %), abnormalities in intestinal morphology (30 %), abnormal body pigmentation (100 %), and shorter body length (47 %) were observed in the 4.03 mg/mL group. Developmental neurotoxicity was characterized by brain apoptosis, damage to the central nervous system (P < 0.01) and peripheral motor neurons (P < 0.0001), and reduced autonomous motor distance (P < 0.01). Transcriptomic analysis revealed that c3a.1, c5, fga, fgb, and fgg were upregulated in the complement and coagulation cascades, and the mRNA levels of c3a.1, dusp1, slc2a6, purba, and klf9 were found upregulated in PCR.CONCLUSIONAcute perchlorate exposure on zebrafish larvae caused various developmental toxicity in a concentration-dependent manner, notably neurotoxicity. We believed the immunological and inflammatory responses were involved.

Unravelling the developmental toxicity of heavy metals using zebrafish as a model: a narrative review

Developmental toxicity is the disruption of an organism's normal development which may occur in either the parent before conception or in the growing creature itself. Zebrafish (Danio rerio) are being employed as effective vertebrate models to evaluate the safety and toxicity of chemicals because they can breed multiple times in a year so we can observe the toxic effects in the next generation and their development mental stages can be observed and define clearly because their 1 cell stage to prime stage is transparent so we can observe the development of every organ also they have nearly about 80% genetic similarity with humans and shares the similar neuromodulatory structure along with multiple neurotransmitter. The recent research endeavours to examine the harmful outcome of various heavy metals such as cadmium, chromium, nickel, arsenic, lead, mercury, bismuth, iron, manganese, and thallium along with microplastics on zebrafish embryos when subjected to environmentally acceptable levels of every single metal in addition to co-exposure at various points in time. These heavy metals can alter the mRNA expression levels, increase the reactive oxygen species (ROS) generation, decrease antioxidant expression, damage neuronal function, alter neurotransmitter release, alter the expression of several apoptotic proteins, interfere with the different signalling pathways, decrease heat rates, increase malformations like - pericardial oedema, heart oedema, reduce in length tail bending abnormal formation in fins. Thereafter we concluded that due to its involvement in the food chain, it also causes severe effects on human beings.

Midostaurin-associated perimyocarditis: a case report of severe cardiotoxicity of novel targeted treatments for acute myeloid leukaemia and review of the literature

AbstractBackgroundMidostaurin is a multikinase inhibitor for the treatment of Fms-like tyrosine 3 (FLT3)-mutated acute myeloid leukaemia (AML). Cardiac adverse events like QTc-prolongation, pericardial effusion, and congestive heart failure have been described. Inflammatory diseases associated with midostaurin are rarely reported.Case summaryA 24-year-old man with newly diagnosed AML and FLT3-ITD mutation was treated with intensive remission-induction chemotherapy and midostaurin. After 5 days of midostaurin, the patient reported severe focal chest pain. Due to laboratory evidence of acute myocardial cell damage, coronary macroangiopathy and pulmonary artery embolism were ruled out via computed tomography. Cardiovascular magnetic resonance showed evidence for active perimyocarditis with myocardial oedema and late gadolinium of the basal, midventricular, and apical lateral wall of the left ventricle. Therapeutic drug monitoring did not reveal excessive midostaurin plasma levels, and hence, initially suspected drug interaction with posaconazole administered for antifungal prophylaxis was considered less likely to be causative. After discontinuing midostaurin, clinical signs of perimyocarditis improved. During continued high-dose cytarabine therapy, no further cardiac events occurred. It was concluded that perimyocarditis was an adverse effect of midostaurin since the inhibition of FTL3 may have led to a loss of cardiomyocyte protective capacity against oxidative stress-induced apoptosis, as previously described in vitro.DiscussionIn addition to the most frequently reported non-cardiac adverse effects of midostaurin, serious cardiotoxic complications appear to occur and may require discontinuation of therapy. This case highlights the importance of interdisciplinary work-up of a cardio-oncology pathway even in presumably low-risk patients and particularly in the context of rare cases of cardiotoxicity in novel cancer treatments.
