This clinical study will evaluate the safety and effectiveness of Gastrin treatment with islet transplantation to help patients with difficult to control type 1 diabetes make insulin again and improve blood sugar control.
This study involves two investigational (experimental) products not yet approved by the U.S. Food and Drug Administration (FDA) as a treatment for any disease:
1. Human allogenic islet cells (islet cells from a deceased, unrelated human donor)
2. Gastrin-17 (Gastrin) - a hormone secreted by the gut

Islet Transplantation Using a T-Cell Depleting Immunosuppression Induction Regimen

City of Hope National Medical Center, located in Duarte, CA, is hosting a clinical study on islet cell transplantation, an experimental procedure being evaluated as a treatment for patients with type 1 diabetes. Islet cell transplantation involves taking insulin-producing cells from organ donors and transplanting them into the liver of a patient with diabetes. Once transplanted, the islets produce insulin, which can improve blood sugar control and eliminate the need to inject insulin or use an insulin pump.
Anti-thymocyte globulin (ATG) and alemtuzumab (Campath) are anti-rejection medications that work by decreasing a patient's T-cells. T-cells are special white blood cells that recognize and destroy unwanted things like infections but can also attack transplanted cells and organs. Reducing the number of T-cells at the time of transplant may protect islets and improve long-term transplant success. In previous research studies, islet transplantation has been successful in reducing low blood sugar episodes, improving overall blood sugar control, and in some cases, allowing patients with type 1 diabetes to stop taking insulin.
The purpose of this study is to determine if islet cell transplantation using ATG or alemtuzumab, along with additional medications to prevent the body from rejecting the transplanted cells, is a safe and effective treatment for type 1 diabetes. Study participants may receive up to three islet transplants and will be followed for five years to monitor blood sugar control, islet transplant function, and changes in quality of life.

An Open Label, Sequential Multi-Center Multi Dose Study Of G17T Immunogen In Combination With Cisplatin (CDDP) And 5-Fluorouracil (5-FU) In Subjects With Metastatic Or Locally Recurrent Gastric Or Gastroesophageal Cancer Previously Untreated With Chemotherapy For Advanced Disease (Stage IV)

RATIONALE: Vaccines may make the body build an immune response to kill tumor cells. Drugs used in chemotherapy use different ways to stop tumor cells from dividing so they stop growing or die. Combining vaccine therapy with chemotherapy may kill more tumor cells.
PURPOSE: Phase II trial to study the effectiveness of combining vaccine therapy and chemotherapy in treating patients who have metastatic or locally recurrent stomach cancer or esophageal cancer.

Unveiling the novel role of spermidine in leaf senescence: A study of eukaryotic translation factor 5A-independent and dependent mechanisms

Senescence is a crucial and highly active process in plants, optimising resource allocation and promoting phenotypic plasticity under restricted conditions. It involves global metabolic reprogramming for the organised disintegration and remobilization of resources. Polyamines (PAs) are polycationic biogenic amines prevalent in all eukaryotes and are necessary for cell survival. The commonly used PAs in plants include putrescine, spermidine, and spermine. Notably, the leaf's expression of S-adenosylmethionine decarboxylase and spermidine synthase gene family transcripts significantly changes during senescence. This suggests these genes are critical in spermidine metabolism and may condition metabolic reprogramming. One key role of spermidine in eukaryotes is to provide the 4-aminobutyl group for the posttranslational modification of lysine in eukaryotic translation factor 5A (eIF5A). This modification is catalysed by two sequential enzymatic steps leading to the activation of eIF5A by converting lysine to the unusual amino acid hypusine. Although eIF5A is well characterised to be involved in the translation of proline-rich repeat proteins and other hard-to-read motifs, the biological role of eIF5A has recently been clarified only in mammals. It could be better described at the plant functional level. The expression patterns of eIF5A isoforms and genes encoding machinery responsible for hypusination, differ between induced and developmental leaf senescence. In this paper, we summarise the existing knowledge on spermidine-dependent senescence control mechanisms in plants, raising the possibility that spermidine could be an element of a biological switch controlling the onset of a different type of senescence in an eIF5A-independent and dependent manner.

Dynamic changes in proanthocyanidin composition, biosynthesis, and histochemistry in spine grape (Vitis davidii Foëx) tissues during berry development

Abstract:

BACKGROUND:

Spine grapes are widely cultivated in southern China because of their strong adaptability to hot and humid climates. As a wild species native to China, spine grape (Vitis davidii Foëx) was studied as a resource of proanthocyanidins (PAs). PA composition, biosynthesis, and histochemistry in different tissues (skins, seeds, and stems) during berry development were analyzed in this study.

RESULTS:

The findings revealed that PA accumulation occurred in concurrence with flowering and was completed by veraison. High‐performance liquid chromatographic results showed that the epicatechin type was the most dominant. The skins were more likely to accumulate PA polymers. Reverse transcription–quantitative polymerase chain reaction analysis revealed that the expression levels of structural genes (flavonoid‐3′‐hydroxylase, flavonoid‐3′5′‐hydroxylase, dihydroflavonol 4‐reductase, leucoanthocyanidin reductase, and leucoanthocyanidin dioxygenase) were positively associated with PA dynamic changes. Histochemical results revealed that PAs in skins were mainly found in the hypodermis of the exocarp, PAs in seeds were mainly found in the middle layer of the outer integument of the testa, and PAs in stems were mainly found in the phloem.

CONCLUSION:

This study provides a clear understanding of the spatial and temporal accumulation of PAs in spine grape, and forms a basis for the analysis of structural profiles and synthesis of PAs and their biological effects. © 2024 Society of Chemical Industry.

Design of High‐Performance Infrared Nonlinear Optical PAs₃S₃ with Perfectly Aligned Polar Molecular Cage via a Bipolar‐Axis‐Symmetry Coupling Strategy

Abstract:

Strong polar molecular cages have recently emerged as novel functional building units for high‐performance infrared nonlinear optical (IR NLO) crystals. However, these highly polar molecular cages often arrange themselves in a way that cancels out their polarity, leading to a more energetically stable state. As a result, most cage crystal formations tend to crystallize in centrosymmetric space groups, which conflicts with the primary requirement for NLO crystals. Herein, we address the challenge of polar molecular cage arrangement through bipolar‐axis‐symmetry coupling strategy, utilizing classical NLO parent compounds. By substituting the C3v symmetric [B3O6] groups with polar C3v symmetric [PAs3S3] cages within the β‐BBO polar aixs lattice, we successfully synthesized a new compound, PAs3S3 (PAS), which exhibits a consistent arrangement of polar molecular cages — crucial for maximizing NLO performance. Additionally, due to the non‐covalent interactions among [PAs3S3] polar molecular cages, PAS demonstrates an unexpectedly strong second harmonic generation (SHG) about 8 times that of AgGaS2, along with a significant band gap of 2.75 eV. Furthermore, PAS exhibits remarkable stability against air and moisture. These findings validate our design strategy and position PAS as a promising candidate for applications in IR NLO crystals.
