Medication-related osteonecrosis of the jaw (MRONJ) occurs after taking bisphosphonates or targeted therapies. It leads to a significant decrease in quality of life with pain, eating and chewing disorders, and malnutrition. Current treatments are only partially effective. PENTO (pentoxifylline and tocopherol) has been shown to be effective in maxillary osteoradionecrosis. The objective of this study is to evaluate the proportion of bone recovery in patients receiving PENTO in MRONJ at 12 months.

Untargeted metabolomic LC-HRMS combined with chemometric reveal metabolites change from sorghum flakes affected by food processing

A non-targeted UHPLC-HRMS metabolomics approach combined with chemometric anal. was employed to assess the change in metabolite profiles of sorghum flakes based on food processing techniques.The result showed that fatty acids were the most abundant metabolites, followed by lipids, amino acids, alkaloids, amines, and polyphenols, representing key metabolic markers influenced by food processing in sorghum flakes.Specific metabolites, including 15S-Hydroxyeicosatrienoic acid (15S-HETrE), terminaline, vitamin E acetate, furan fatty acid F6, all-trans-retinal, and fingolimod, exhibited increased concentrations during oven processing compared to raw sorghum, likely due to their thermal stability.This study provides valuable insights into effect of food processing effect on metabolites profiles.

Isotope tracing-assisted chip-based solid-phase extraction mass spectrometry for monitoring metabolic changes and vitamin D3 regulation in cells

Cellular metabolism is a dynamic and essential process, with alterations in metabolic pathways serving as hallmark features of cancer. In this study, we developed a chip-based solid-phase extraction mass spectrometry (Chip-SPE-MS) platform for high-sensitivity, high-throughput analysis of cellular metabolites and real-time tracking of metabolic fluxes. The system achieved detection limits ranging from 0.10 to 9.43 μmol/mL for various amino acids and organic acids, with excellent linearity (r ≥ 0.992). By incorporating isotope tracing, the platform enabled derivatization-free, real-time monitoring of ¹³C-labeled metabolites, such as lactic acid. Our analysis revealed significant metabolic differences between normal (L02) and cancerous (HepG2, HCT116) cells, including enhanced glycolytic activity and elevated lactate production in cancer cells. Furthermore, treatment with 1,25-dihydroxyvitamin D3 was shown to suppress glucose uptake and modulate metabolic activity in HCT116 cells, highlighting the regulatory effects of vitamin D3 on cancer metabolism. This study not only provides novel insights into the metabolic reprogramming associated with cancer but also demonstrates the potential of the Chip-SPE-MS platform as a powerful tool for real-time monitoring of dynamic metabolic processes. The findings have broad implications for cancer therapy and the study of metabolic diseases.

Ground-level ozone (O₃) is well recognized as a secondary air pollutant with detrimental effects on plant growth and biochemistry. In a field study, Andrographis paniculata (King of Bitter) was exposed to ambient O₃ and elevated O₃ (AO + 20 ppb) at three growth stages [45, 90, and 135 days after treatment, (DAT)] using open-top chambers. Elevated O₃ stress negatively impacted plant growth, increased cell damage, and induced foliar injuries. However, elevated O₃ also boosted antioxidant production such as proline, phenol, and enzymatic antioxidants, as well as certain secondary metabolites such as tannins, phytosterols, saponins, and alkaloids. This may enhance the plant's medicinal properties, including compounds limonene dioxide, phytol, palmitic acid, and androstadiene. While, certain metabolites like Citronellol, Khusenol, and tocopherol displayed an adverse reaction under elevated O₃ exposure. The novel detection of acrodiene, squalene, and neophytadiene under O₃ stress emphasizes their medicinal significance. Notably, an important bioactive compound andrographolide in A. paniculata showed increased synthesis under elevated O₃ at 45 and 90 DAT, suggesting that O₃ exposure could enhance the plant's pharmaceutical value.
