Adia Med of Winter Park LLC Chronic Kidney Disease Research Study

The goal of this clinical trial is to learn whether a new regenerative treatment called AdiaVita, made from umbilical cord blood-derived stem cells and exosomes combined with glutathione, is safe and can help improve kidney function in adults with chronic kidney disease (CKD). In this condition, the kidneys gradually lose their ability to filter blood as well as they should. The main questions it aims to answer are whether AdiaVita plus glutathione improves kidney function better than control treatments, as measured by blood tests for estimated glomerular filtration rate (eGFR) and creatinine levels, and whether the treatment is safe with acceptable side effects. Researchers will compare three groups. One group will receive AdiaVita plus glutathione. A second group will receive glutathione plus a placebo for AdiaVita. The third group will receive placebos for both treatments. A placebo looks like the real treatment but contains no active ingredients. This will help determine if the full treatment works better than the controls. Approximately 100 adults aged 18 to 80 with stage 2 to 4 chronic kidney disease may participate. Participants will be randomly assigned to one of the three treatment groups. They will receive monthly intravenous infusions at the clinic for the first three months and apply a skin spray twice daily at home during that period. The study lasts 12 months total for each participant, with regular visits for blood tests, physical exams, and safety monitoring. Certain participants in the control groups may switch to the active AdiaVita treatment after three months if they meet safety criteria. This is a single-blind study, meaning participants will not know which treatment they receive. Participant safety is closely monitored by the research team and an independent board throughout the study.

Adia MED of Winter Park LLC Autism Spectrum Disorder Research Study

This 24-month study is testing whether adding AdiaVita, an umbilical cord blood-derived stem cell and exosome product, to glutathione therapy helps improve autism symptoms in children ages 3-12 more than glutathione alone. Children will be randomly placed into one of two groups for the first three months: one group receives glutathione only, and the other receives glutathione plus monthly intravenous AdiaVita infusions. Both groups also use topical glutathione cream twice daily at home. Autism symptoms will be tracked over two years using the Autism Treatment Evaluation Checklist (ATEC) filled out by parents and by therapists or teachers. Safety, side effects, quality of life, and overall well-being will be closely monitored through regular clinic visits, physical exams, blood tests, and adverse event reporting. After the initial three-month phase, children who received glutathione alone may cross over to receive AdiaVita infusions at no additional cost if safety checks at month 6 are satisfactory. Approximately 100 children with a confirmed autism diagnosis from the Central Florida area will take part. Participation is completely voluntary, and families may withdraw at any time.

CeO2@CA@PEI@dextran improves drug sensitivity in chronic myelogenous Leukemia through scavenging free radical efficiently

The sustained free radicals derived from drug-resistant chronic myeloid leukemia (CML) cells can trigger oxidative stress, thereby reducing cell drug sensitivity and therapeutic efficacy. Our prior work demonstrated that dextran-modified ceria nanoparticles (CeO₂ NPs) exhibit the intrinsic antioxidant capability and are capable of enhancing anti-leukemia therapeutic responses. However, excessive intracellular free radicals trigger the interfacial electron transfer across nano-enzymatic surfaces, disrupting Ce³⁺/Ce⁴⁺ valence equilibrium and compromising their long-term antioxidant performance. Here, we found that CeO₂ NPs modified by citric acid (CA) and polyethyleneimine (PEI) and dextran (CeO₂@CA@PEI@Dextran) provided the long-lasting capacity to scavenge radicals by the aid of electronic buffer effect of CA@PEI, which can buffer the process of electronic gain and loss to maintain the stability of Ce³⁺/Ce⁴⁺ ratio and a long-term performance to scavenge free radicals in vitro and in vivo. CeO₂@CA@PEI@Dextran thus regulated intracellular oxidative stress persistently to improve the effects of doxorubicin in K562-ADR cells, one CML cell line resistant to doxorubicin. In contrast, the regulatory effect of small-molecule antioxidants such as N-acetylcysteine and glutathione in diminishing free radicals is unsustainable and undulant. Consequently, reshaping the redox homeostasis in CML patients by the continuous anti-oxidation mediated by CeO₂@CA@PEI@Dextran may be a potential way to improve the treatment of drug-resistant patients, distinct from the unsustainable anti-oxidation strategies, which presents a novel perspective for clinical drug usage.

Smartphone-assisted colorimetric sensor array based on trimetallic FeMnZn-NC nanozyme for the pattern recognition of antioxidants in fruit

Antioxidants such as ascorbic acid, glutathione, gallic acid, and caffeic acid are crucial for food quality and human health, yet their simultaneous discrimination in complex food matrices remains challenging due to overlapping reactivities. In this study, we developed a robust colorimetric sensor array based on trimetallic FeMnZn‑nitrogen‑carbon (FeMnZn-NC) nanozymes. Synthesized via high-temperature pyrolysis, the introduction of Zn as a sacrificial template generates abundant structural defects and exposes more active sites, resulting in significantly enhanced oxidase-like activity compared to mono- and bimetallic analogues. The nanozymes catalyze the rapid oxidation of TMB and ABTS substrates within 2 min, a process differentially inhibited by distinct antioxidants via varying electron-transfer and chelating mechanisms. Leveraging these unique kinetic "fingerprints," a dual-channel sensor array was constructed to identify ascorbic acid, glutathione, gallic acid, and caffeic acid, exhibiting linear responses over 0-40 μM with detection limits of 0.22, 0.43, 0.21, and 0.14 μM, respectively. The sensor demonstrated excellent anti-interference capability and was effectively applied to identify antioxidants in apples, pears, and kiwifruits. The integration of a smartphone-assisted colorimetric platform enables rapid, on-site detection, highlighting its strong potential for food quality monitoring and nutritional assessment.

Protective role of VitC, Glutathione and Sodium Pyruvate against antibiotic-induced oxidative damage in Escherichia coli

The increasing prevalence of antibiotic tolerance poses a significant challenge in the treatment of bacterial infections. This study investigates the inhibitory effects of commonly used antibiotics- Ciprofloxacin (CFX), Levofloxacin (LVX), and Ampcillin (Amp)-on exponentially growing Escherichia coli DH5α cells, and further explores the modulatory roles of antioxidants including Vitamin C(VitC), Glutathione, and Sodium pyruvate (Na-Pyr). Minimum inhibitory concentrations (MICs) were determined using the microplate Alamar Blue assay (MABA), revealing that fluoroquinolones (CFX and LVX) exhibited highest potency while Amp demonstrated moderate efficacy. Dose-dependent inhibition was observed for each antibiotic, with near-complete suppression of bacterial growth at higher concentrations. Interestingly, co-treatment with antioxidants significantly altered antibiotic performance. VitC enhanced the efficacy of CFX, LVX, and ampicillin. Glutathione increased the activity of CFX and also enhanced LVX and ampicillin efficacy, but primarily at lower concentrations. Na-Pyr showed minimal effect on CFX, while it increased the efficacy of LVX and Amp at lower concentrations. This suggests that these antioxidants may modulate redox homeostasis and amplify reactive oxygen species (ROS)-mediated stress, thereby boosting antibiotic lethality. Collectively, these findings highlight the contribution of oxidative stress to antibiotic action and suggest that co-treatment with specific antioxidants may serve as a promising strategy to improve therapeutic outcomes. This study adds to our understanding of exponentially growing cells susceptibility and antibiotic tolerance, revealing how metabolic and redox-modulating compounds can be leveraged to enhance bacterial killing.
