EAT-UP - Extended Antibiotic Treatment in Chronic UTI Patients; a Phase II Safety and Efficacy Trial

Chronic Urinary Tract Infection (UTI) is a type of UTI where symptoms are constant and occur every day, unlike recurrent UTIs, which come and go with symptom-free breaks in between. Current treatment for chronic UTI within the NHS is based on recommended guidelines for recurrent UTI. The standard approach typically includes one of the following treatments:
* Long-term, prophylactic (low) dose daily antibiotic (where medication is used at low doses to try to prevent symptoms reoccurring).
* Long-term use of a urinary antiseptic (which helps keep your urine bacteria free), called methenamine hippurate.
These often do not work for people with chronic UTI, and symptoms can persist. Moreover, standard urine tests may fail to detect infections, making diagnosis and treatment more challenging.
The EAT-UP trial will investigate whether longer courses of treatment (higher) dose antibiotics combined with methenamine hippurate (a urinary antiseptic) are a more effective treatment at reducing levels of infection and symptoms than standard of care treatments (as described above).

A hybrid supercapacitor containing NiO microflowers as cathode material with enhanced energy storage performance

The NiO microflowers electrode material was synthesized through a simple and low-cost hydrothermal method using hexamethylenetetramine (HMTA) to provide hydroxide ions for the formation of corresponding precursors. On this basis, the effect of HMTA dosage on the morphology and electrochemical performance of nickel oxide was investigated. Two kinds of NiO powdered materials with distinct morphologies were successfully prepared. The NiO microflowers demonstrated a larger specific surface area (SSA, 142.43 m² g^-1) and a higher specific capacity (C_s) of 661.4 C g^-1. While the NiO microspheres possessed smaller SSA of 104.3 m² g^-1 and inferior C_s of 594.6 C g^-1. The NiO materials were employed as the cathode and activated carbon (AC) as the anode to assemble hybrid supercapacitor (HSC), which was expressed as NiO microflowers//AC and NiO microspheres//AC, respectively. Compared with the 53.5 W h kg^-1 energy density (ED) of NiO microspheres//AC, the NiO microflowers//AC achieved a higher ED of 55.4 W h kg^-1. Both HSCs showed over 90 % capacity retention after 4000 cycles at 10 A g^-1, indicating that the two HSCs exhibited excellent cyclic lifespan. It suggests that the NiO electrode materials in this study show promising potential for electrochemical energy storage application. The current synthesis approach demonstrates notable advantages including simplicity, time and energy saving, and may be straightforwardly employed to prepared other electrode materials with controlled morphologies and superior electrochemical performance.

Histone methylation of kidney disease: fact or fantasy?

Histone methylation (HMT), the enzymatic addition of methyl groups to specific histone residues by histone methyltransferases, constitutes a key regulatory mechanism in gene expression and post-translational modulation. Although studies have explored HMT's role in oncogenesis and other organ-specific disorders, HMT is now implicated in the pathogenesis of kidney diseases. A broad spectrum of experimental models, including both in vivo and in vitro systems, has demonstrated the involvement of HMT alterations in diverse renal pathologies such as acute kidney injury, renal fibrosis, diabetic nephropathy, lupus nephritis, polycystic kidney disease, kidney stones, renal cell carcinoma, and immunoglobulin A nephropathy. Targeted modulation of HMT has been associated with attenuated disease progression across these conditions. This review synthesizes current insights into HMT's mechanistic roles in renal pathology and delineates its therapeutic potential as a strategic intervention in kidney disease management.

Effect of sequential hydrothermal treatments on the physicochemical properties and in vitro digestibility of bitter vetch (vicia ervilia) starch

This study evaluated the effects of single (heat-moisture treatment (HMT, 120 °C/24 h, 30 % moisture) and annealing (ANN, 50 °C/24 h, 70 % moisture)) and sequential (HMT-ANN and ANN-HMT) hydrothermal modifications on the physicochemical and digestibility properties of bitter vetch starch (BVS). The most significant reduction in peak and setback viscosities was observed in HMT sample, decreasing from 3278 ± 33 cP to 1467 ± 47 cP and from 3199 ± 79 cP to 406 ± 10 cP, respectively, compared to the native starch. DSC experiment confirmed elevated gelatinization temperatures, with onset (T_o) increasing from 59.54 ± 0.08 °C (native) to 69.35 ± 0.01 °C (HMT-ANN), accompanied by higher enthalpy (ΔH) values, from 7.12 ± 0.05 J/g to 11.40 ± 0.13 J/g. XRD and FTIR analyses indicated increased crystallinity (from 33.51 to 41.78 %) and short-range molecular order (1047/1022 ratio from 0.9812 to 0.9885), whereas SEM images of starch gels revealed primarily morphological changes. Gel hardness was notably reduced following HMT (222.1 ± 9.1 g) and HMT-ANN (253.6 ± 9.3 g), suggesting that dense crystallinity and limited chain mobility hindered the formation of firm gels. Importantly, in vitro digestibility assays showed that HMT and HMT-ANN treatments increased resistant and slowly digestible starch fractions by approximately 1.5- and 4-fold, respectively, indicating potential for low-glycemic food applications. These findings highlight the promising role of hydrothermal treatments in tailoring the functionality of BVS for clean-label, health-promoting food systems.

Zydus Methenamine Hippurate tablet gets FDA final nod

New Delhi: Zydus Lifesciences announced its final approval from the US Food and Drug Administration to manufacture and market Methenamine Hippurate tablet in the country’s drug market. The drug is indicated for prophylactic or suppressive treatment of frequently recurring urinary tract infections when long-term therapy is considered necessary. Methenamine Hippurate tablets is a generic drug and the company will manufacture the drugs at its Ahmedabad facility . As per IQVIA analysis in the last twelve month period ending January 2025, Methenamine Hippurate tablets had reported net sales of $32.6 million in the United States. Govt bans 156 medicines including antibiotics, painkillers, multivitamins: Here is the full list

New Delhi: In a significant move aimed at safeguarding public health, the Indian government has banned 156 fixed-dose combination (FDC) drugs, including a variety of antibiotics, painkillers, multivitamins, and medications for treating fever and hypertension. This decision marks the largest crackdown on FDCs since 2016, when 344 such drugs were prohibited. The Union Ministry of Health and Family Welfare issued a gazette notification on Thursday, officially prohibiting the manufacture, sale, and distribution of the banned FDCs. The ban was based on the recommendations of an expert panel that reviewed a total of 324 FDCs. Impact on Pharmaceutical Industry The banned FDCs include popular combinations such as mefenamic acid and paracetamol injections, commonly used for pain relief and fever, and omeprazole magnesium with dicyclomine HCl, used to treat abdominal pain. Hormone-Modulating Therapy May Reduce Dementia Risk in Breast Cancer

THURSDAY, July 25, 2024 -- For women with breast cancer, hormone-modulating therapy (HMT) is associated with reduced risk of Alzheimer disease and related dementias (ADRD), according to a study published online July 16 in JAMA Network Open . Chao Cai, Ph.D., from the University of South Carolina in Columbia, and colleagues examined the association between HMT for breast cancer treatment and the risk of developing ADRD among women aged 65 years or older in a cohort study. Of 18,808 women aged 65 years and older diagnosed with breast cancer between 2007 and 2009, 65.7 and 34.3 percent received and did not receive HMT, respectively, within three years after diagnosis. The researchers found that during an average of 12 years of follow-up, 23.7 and 27.9 percent of HMT users and non-HMT users developed ADRD. Overall, HMT was associated with a reduced relative risk of ADRD (hazard ratio [HR], 0.93). The association varied by race and decreased with age. The greatest reduction in ADRD risk associated with HMT was seen for women aged 65 to 74 years who self-identified as Black (HR, 0.76); among women aged 75 years or older, the association was attenuated (HR, 0.81). The relative risk reduction was lower for women aged 65 to 74 years who self-identified as White (HR, 0.89) and was no longer significant for those aged 75 years or older. No significant association was seen for HMT and ADRD for other races. "Further research is warranted to validate the observed associations in diverse populations and to elucidate the mechanisms underlying our observations," the authors write.
