Season-dependent variation in the contents of glucosinolates and S-methyl-l-cysteine sulfoxide and their hydrolysis in Brassica oleracea

Brassica vegetables contain glucosinolates and S-methyl-l-cysteine sulfoxide, which can be enzymatically hydrolyzed to form bioactive compounds. Glucosinolate hydrolysis can result in formation of health-promoting isothiocyanates, however, often less desirable nitriles and epithionitriles are formed due to presence of specifier proteins. Also, S-methyl-l-cysteine sulfoxide yields beneficial volatile organosulfur compounds (VOSC), such as S-methyl methanethiosulfinate. To optimize the nutritional value of Brassica oleracea vegetables, the outcome of these hydrolysis pathways was monitored over the harvest seasons of different cultivars in three consecutive years. Strong seasonal shifts in glucosinolate hydrolysis were observed only in red cabbages. They formed up to 40-fold more isothiocyanates in summer than in autumn, when nitriles and epithionitriles increased due to the induction of epithiospecifier proteins. Further, higher VOSC levels were found in autumn red cabbage. By elucidating the impact of abiotic growth factors on the regulation of the hydrolyses, the health value of Brassica vegetables can be improved.

Outcomes of percutaneous endoscopic versus endoscopic transmural necrosectomy for necrotizing pancreatitis: A propensity score-matched study

BACKGROUNDFew published studies exist that compare the outcomes of different endoscopic necrosectomy methods for necrotizing pancreatitis (NP). We compared the safety and efficacy of percutaneous versus transmural endoscopic necrosectomy for NP patients.METHODSIn this retrospective cohort study, we analyzed adult NP patients who underwent either percutaneous endoscopic necrosectomy (PEN) or endoscopic transmural necrosectomy (ETN), and compared safety and efficacy between the two groups. Propensity score-matched analysis and multivariable logistic regression analysis were conducted.RESULTSA total of 280 patients were enrolled, among which 142 underwent PEN and 138 underwent ETN. There were differences in baseline characteristics between the two groups, including body mass index, C-reactive protein, systemic inflammatory response syndrome score. The incidences of sepsis, respiratory failure, and intensive care unit stay were higher among patients who underwent PEN than those who underwent ETN (all P < 0.01). Ninety-one pairs were matched with comparable baseline characteristics and severity. The incidence of postoperative complications, open surgery, clinical success, radiological success, collection recurrence, and reintervention were not significantly different between the ETN group and PEN group (all P > 0.05). Multivariate analysis also showed that the approaches (PEN vs ETN) was not associated with postoperative complications or mortality.CONCLUSIONSIn real world setting, sicker patients tend to be more effectively managed through PEN compared to ETN. PEN demonstrates comparable efficacy and safety to ETN in the treatment of NP patients.

Combined Solid‐Phase Extraction of Organic, Organic Peroxide, and Inorganic Explosives for Post‐Blast Residue Analysis

ABSTRACTPost‐blast explosives residue may contain organic and/or inorganic explosives. Current methods employed involve splitting the sample swab in two for syringe extraction for each explosive type (acetone for organics, water for inorganics). A comprehensive solid‐phase extraction (SPE) trace sample clean‐up procedure for the combined recovery of 12 organic, 2 organic peroxide, and 15 inorganic analytes from explosives residue on cotton swabs is demonstrated and validated. By keeping the sample whole and using a single extraction process, the proposed method saves time, provides a more representative sample, and increases the possibility of detecting explosives that might be present on collection swabs. Thus, recoveries are improved for one class of explosives without sacrificing recoveries of another analyte or explosives class in the process. Extractions were demonstrated by syringe filtration and SPE from cotton balls (CBs) and cotton‐tipped applicators (CAs). For SPE of both matrices, organic explosives (nitramines, nitrate esters, nitroalkanes, and nitroaromatics) had a limit of detection (LOD) of 500 ppb or lower, organic peroxides had higher LODs at 5 ppm or lower, and most inorganic anions and cations had LODs at 1 ppm or lower. Application to 32 post‐blast samples, 9 burned powders simulating post‐blast, and 1 pre‐blast sample successfully demonstrated the SPE method with both cotton matrices in a comparison study of the two extraction procedures. SPE resulted in a lower false screen rate (number of compounds that met the screening criteria, but not confirmed present) when compared to syringe extraction.
