Eisai has stopped the Phase 3 clinical development of an adjunctive treatment for Dravet syndrome, a spokesperson confirmed to Endpoints News on Friday afternoon. The Tokyo drugmaker terminated the late-stage trial of lorcaserin this week, according to an update to the federal clinical trials database. The medicine, which has FDA orphan drug designation, was still listed on Eisai’s public pipeline as of Nov. 8. Dravet, a rare disorder, leads to epilepsy and affects people starting in infancy. “As other FDA-approved therapies have become available to treat this complex condition, recruiting patients for an investigational therapy has been exceedingly challenging,” the Eisai spokesperson said in an email. They said the decision wasn’t related to safety issues. Eisai got the oral drug from Arena Pharmaceuticals and marketed it as Belviq for obesity. But in 2020, the FDA requested that Eisai pull the drug after concerns about cancer outweighed its benefits. The drug is a 5-HT2C agonist that selectively activates serotonin 2C receptors in the brain. The goal is to suppress seizures in patients with Dravet syndrome. Another drugmaker developing a 5-HT2C superagonist, Longboard Pharmaceuticals, entered Phase 3 in Dravet patients this fall. Lundbeck is buying Longboard for $2.6 billion in cash. Bright Minds Biosciences is also developing a related drug and is in mid-stage testing in rare epilepsies, with preclinical work in obesity, as well. Stoke Therapeutics, Jazz Pharmaceuticals and other companies are also working in Dravet. Takeda’s drug failed a Phase 3 this summer.
