Inflammation, immune responses, cell migration, autophagy and cell signaling are all important physiol. functions where members of the protein family galectins play an important role. Members of this family of proteins, in particular galectin 3, have also been associated with disease pathol. of for example fibrosis and cancer. Fibrosis targets different organs such as lung, liver, heart and skin and upon progression eventually results in organ failure. In industrialized nations, fibrosis has been estimated to account for 45% of all deaths which emphasizes the need to develop treatments targeting different organs.<br/>Our first small mol. galectin 3 inhibitor in clinic, TD139 is being developed as an inhaled treatment of idiopathic pulmonary fibrosis (IPF). This compound recently entered Phase IIb trials after a successful phase 1/2a trial where TD139 had significant effect on biomarkers typical for IPF. TD139 is not suitable for oral delivery and therefore we have developed a new series of high affinity and orally available galectin 3 inhibitors. This lecture will discuss the pharmacol. of galectin 3 and the discovery process leading up to selection of GB1211, the first orally available galectin inhibitor taken to clinic (phase 1a ended Jan. 2020) and the progress in clinic so far.

Design of ligands targeting carbohydrate binding sites: Galectin 3

Designing small high affinity lectin inhibitors starting from natural saccharides is a major challenge due to their polarity and that the matching lectin binding sites not only are polar but also shallow. The lectin/ligand interactions are often made up by complex hydrogen networks which in some cases are strengthened by other interactions such as CH-π stacking between the ligand carbohydrate skeleton and aromatic aminoacids. Therefore, natural mono- and disaccharides in general have low binding affinites in the μM-mM range. Polar ligands like saccharides in general also have low passive uptake over the intestine and often rapid renal or biliary excretion. Despite these challenges several ligands targeting carbohydrate binding sites of different target classes have been discovered and developed into medicines. Examples are the heparin mimetic fondaparinux, the glifozins which are glucose transporter inhibitors (SGLT2), inhibitors of the uropathic E. coliadhesin FimH,influenza neuramnidase inhibitors zanamivir and oseltamivir, selectin inhibitors, siglec inhibitors and the iminosugar miglastat, which last year was approved by FDA for treatment of Fabry disease. We have developed small high affinity carbohydrate-based ligands targeting galectin 3, a target involved in the pathol. of inflammation and fibrosis. Our first inhibitor TD139 is currently in phase IIb trials, being developed as an inhaled treatment of idiopatic pulmonary fibrosis (IPF). Recently we discovered an orally available galectin 3 inhibitor GB1211 which has been taken into phase 1 clin. trials in healthy volunteers targeting non-alc. steatohepatitis (NASH). This presentation will discuss learnings from the design of ligands targeting carbohydrate binding sites with examples from both our own galectin research and literature.

NextCure hires Celgene vet as CMO; José Vega leaves Merck to become Moderna's chief safety officer

NextCure has found its new CMO after the previous executive resigned last August. Han Myint, a biopharma and academia veteran of more than 20 years, joined the company Thursday after serving as CMO at NexImmune for a little over a year. Myint had overseen the company’s Phase I/II trial for a cell therapy in acute myeloid leukemia before hopping over to NextCure. “Han’s strong industry track record is highlighted by his involvement in the development of multiple U.S. Food and Drug approved products,” CEO Michael Richman said in a statement. At NextCure, Myint will turn his attention toward the company’s two lead programs: NC318, an immunotherapy targeting Siglec-15, and NC410, a recombinant LAIR-2 fusion protein designed to block immune suppression mediated by the immune modulator LAIR-1. Before joining NexImmune, Myint had a senior role at Celgene, serving as VP of global medical affairs and the myeloid diseases lead for more than six years. He helped shepherd through a number of FDA-approved products during his time there, NextCure said. Myint had previously worked at the University of Colorado-Denver between 2005 and 2012 as a professor and as the director of hematological malignancies and stem cell transplant program, building the program out. David Kessler resigns from the Biden admin, marking the unofficial end of Operation Warp Speed The White House David Kessler, M.D., is resigning as chief scientific officer of the Biden administration’s COVID response, marking the end of Operation Warp Speed, the famed federal effort to develop and administer COVID vaccines. His departure is a tangible checkpoint in the COVID-19 pandemic, as COVID vaccinations transition from being organized and overseen by the federal government to the private sector. In an exit interview with The Wall Street Journal, Kessler said, “There has never been a successful vaccine campaign as large as this.” Kessler’s resignation marks the end of his latest chapter in public service, which began in 1990 when President George H.W. Bush appointed him to be commissioner of the FDA. He served in the role until 1997 when he stepped down, later becoming dean of the Yale School of Medicine. Kessler’s role as chief scientific officer of the pandemic response was much different than that of his predecessors in the prior administration. Whereas leaders of Operation Warp Speed worked on expediting development of COVID vaccines, Kessler was in charge of administration. And over the last three years, more than 650 million doses have been administered. And while new infections and hospitalizations have remained lower than prior highs, efforts to roll out new booster shots have proved more difficult. Tweet Novocure shakes up C-suite as tumor-treating field pipeline expands Novocure Cancer-treating medical device company Novocure is shaking up its executive team as the company’s chief medical officer steps down following a “review of Novocure’s future needs.” The decision by Ely Benaim, M.D., comes days after the company posted a positive readout of a phase 3 trial testing its tumor treating fields in conjunction with standard therapy as a treatment for non-small cell lung cancer. His move marks the end of a three-and-a-half year career at AstraZeneca where he most notably oversaw development of the company’s COVID-19 vaccine—built in collaboration with the University of Oxford—and its FDA-approved treatment, Evusheld. The work earned him a British order of chivalry. Prior to joining AstraZeneca in 2019, Marshall spent more than 16 years at GSK, rising through the ranks from director of clinical pharmacology to medicines development leader. He left GSK in 2019 to become chief medical officer at Galecto Biotech, a role he kept for just more than a year. Juvenescence dubs itself as an anti-aging therapeutics company, a broad aspiration that centers on developing therapies via a variety of modalities like regenerative medicine or metabolism revival. The company launched in 2017 and raised $100 million two years later to advance its programs. The company’s lead asset, LyGenesis, is in a phase 2 trial to treat end-stage liver disease.
