Recent Pipeline Updates VYN201, a locally-administered pan-BD BET inhibitor: Selected formulation for Phase 2b trial – Following extensive testing, VYNE plans to evaluate VYN201 in a once-daily gel formulation for the upcoming Phase 2b trial in patients with nonsegmental vitiligo. The Company believes the gel formulation may provide additional clinical benefits based on improved dermal penetration properties compared to the ointment that was evaluated in the Phase 1b trial. Engaged experienced contract research organization (“CRO”) – VYNE has engaged a CRO with extensive experience conducting Phase 2 and Phase 3 vitiligo trials for its upcoming Phase 2b trial. Trial startup activities are ongoing with patient enrollment expected to begin in the second quarter of 2024. Top line results from the 24-week double-blind portion of the Phase 2b trial are anticipated in mid-2025. Submitted Phase 2b trial design to FDA – The Phase 2b trial is anticipated to be a 24-week randomized, double-blind, vehicle-controlled trial with a separate active treatment extension phase to 52 weeks. Pending the U.S. Food and Drug Administration's (“FDA”) acceptance of the protocol, the trial will evaluate three or four arms (including vehicle) of once-daily treatment with VYN201 gel, with each arm enrolling between 40 and 50 patients with active or stable nonsegmental vitiligo. The primary efficacy endpoint of the trial will be an evaluation of the proportion of subjects achieving FVASI50 at week 24 compared to vehicle. Announced positive biomarker data from Phase 1b trial – Earlier this year, VYNE announced that exploratory data for the 1.0% and 2.0% cohorts from the Phase 1b trial showed that VYN201 treatment demonstrated biological activity and a positive effect on certain key biomarkers relevant to vitiligo disease severity and progression. This data supports VYN201’s rapid onset of action and previously announced positive clinical results. VYN202, an oral small molecule BD2-selective BET inhibitor: Completed additional nonclinical study in support of IND – VYNE submitted an Investigational New Drug application ("IND") for VYN202 to the FDA in December 2023. Before commencing a Phase 1a clinical trial, the FDA requested data from an additional nonclinical study. VYNE recently completed this study and achieved preliminary results consistent with its expectations at the outset of the study. This additional study evaluated doses of VYN202 within the expected clinical therapeutic window of the BET target. Consistent with previous nonclinical studies for VYN202, there were no apparent off-target toxicities observed. VYNE plans to submit the requested nonclinical information to the FDA by the end of the first quarter of 2024. Phase 1a trial expected to start in Q2 – Assuming the IND is cleared by the FDA, VYNE expects to initiate its Phase 1a single ascending dose/multiple ascending dose (SAD/MAD) trial in healthy volunteers in the second quarter of 2024, with top line results from the trial anticipated in the second half of 2024. If the Phase 1a portion of the trial is successfully completed, VYNE plans to initiate Phase 1b trials in subjects with moderate-to-severe plaque psoriasis and moderate-to-severe adult-onset rheumatoid arthritis, with top line results anticipated in the second half of 2025. Financial Results for the Year Ended December 31, 2023 Revenues. Revenues for the year ended December 31, 2023 totaled $0.4 million compared to $0.5 million for the year ended December 31, 2022, consisting of royalty revenue from sales of Finacea foam by LEO Pharma, to whom VYNE previously licensed the rights to that product. Research and development expenses. VYNE’s research and development expenses for the year ended December 31, 2023 were $16.3 million, representing a decrease of $2.1 million, or 11.3%, as compared to $18.4 million for the year ended December 31, 2022. Both newly approved and well-established treatments recommended ROSEMONT, Ill., Jan. 31, 2024 /PRNewswire/ -- Acne is the most common skin condition in the United States, affecting nearly 50 million Americans each year, with symptoms usually beginning in puberty. Acne affects 85% of adolescents and can also often begin or continue in adulthood, especially in women. To help ensure that people with acne get the best possible treatment, the American Academy of Dermatology (AAD) has published updated guidelines of care for the management of acne in adults, adolescents, and children over the age of 9 in the Journal of the American Academy of Dermatology. Continue Reading "These guidelines provide important updates to the 2016 AAD acne guidelines, including discussion of new topical medications, which are directly applied to the skin, and systemic treatments, which are taken by mouth," said board-certified dermatologist John S. Barbieri, MD, MBA, FAAD, co-chair of the AAD's Acne Guideline Workgroup. "This update provides evidence-based recommendations for dermatologists and other clinicians caring for patients with acne." Strong recommendations were made for the use of the following: Topical benzoyl peroxide to reduce the amount of acne-causing bacteria on the skin. Topical retinoids (e.g. adapalene, tretinoin, tazarotene, trifarotene) to help unclog pores and reduce inflammation. Oral antibiotics, such as oral doxycycline, and topical antibiotics to reduce the amount of acne-causing bacteria on the skin and lessen inflammation. Combinations of topical benzoyl peroxide, retinoids, or the above antibiotics. Employing best practices in guideline development, the multidisciplinary workgroup also issued 5 good practice statements. When managing acne with topical medications, the guidelines recommend combining multiple different treatment types, as this can lead to better results. The guidelines recommend limiting use of oral antibiotics when possible to reduce the development of antibiotic resistance and other antibiotic-associated complications. It is recommended that topical and oral antibiotics are used simultaneously with benzoyl peroxide to prevent the development of antibiotic resistance. For patients with larger acne bumps, the guidelines recommend injectable corticosteroids as a potential option for more rapid relief of inflammation and pain. For patients with severe acne or for patients who have failed standard treatment with oral or topical therapy, the guidelines recommend isotretinoin. Additionally, the guidelines provide conditional recommendations. Conditional recommendations apply to most patients, but the most appropriate action may differ depending on individual patient factors. Topical clascoterone, which addresses hormonal causes of acne. Topical salicylic acid to unclog pores and exfoliate the skin. Topical azelaic acid to unclog pores, kill bacteria, and fade dark spots that may continue when an acne spot clears. Oral minocycline or sarecycline to reduce the amount of acne-causing bacteria on the skin and lessen inflammation. Hormonal therapies such as combined oral contraceptives or spironolactone to address hormonal causes of acne. Available evidence was insufficient to develop recommendations for procedures such as chemical peels, laser and light-based devices, microneedling, as well as for dietary changes, or alternative therapies such as vitamins or plant-based products. Additionally, a conditional recommendation against adding broadband light, intense pulsed light, to adapalene 0.3% gel.
