Acute exogenous oestradiol augments endothelial and smooth muscle vasodilatory responsiveness in premenopausal but not postmenopausal females

AbstractMenopause is associated with vascular dysfunction. During the menopausal transition, endogenous oestradiol concentrations diminish. Oestradiol is vasoprotective because it has direct and indirect effects on the vasculature. The present study aimed to determine the effect of acute exogenous oestradiol on endothelium‐dependent, endothelium independent and β2‐adrenergic receptor‐induced vasodilatation in females. Forearm blood flow (venous occlusion plethysmography) was measured during brachial intraarterial infusions of ACh (endothelium‐dependent agonist), sodium nitroprusside (endothelium independent agonist) and terbutaline (β2‐adrenergic receptor agonist) with and without concurrent infusion of 17β‐oestradiol. Nine young premenopausal (age: 26 ± 4 years) and nine postmenopausal (PM, age: 58 ± 4 years, 8 ± 1 years post‐menopause) females completed the study. Concurrent oestradiol infusion augmented the vasodilatory response to ACh, sodium nitroprusside and terbutaline in young premenopausal (all P < 0.05) but not older postmenopausal (all P > 0.05), females. Local infusion of exogenous 17β‐oestradiol augmented endothelial and smooth muscle microvascular vasodilatation in premenopausal but not postmenopausal, females.

Because oestradiol has vasoprotective effects, the menopause‐associated drop in oestradiol concentrations is hypothesized to contribute to vascular dysfunction during the menopause transition.

Increasing chemical coverage, accuracy, and reproducibility of the processing method for polar organic chemical integrative samplers

AbstractDespite their diffusion in research studies, passive samplers are rarely used in regulatory applications. To expand the employment of passive samplers in regulatory environmental studies, standardized procedures for processing each sampler type should be proposed and accepted, but currently, each study develops its own protocol based on previous knowledge and specific needs. In this work, six identical polar organic chemical integrative samplers in seawater were deployed to understand the importance of the sorbent transfer method prior to the elution step. A common “wet transfer” with ultra-pure water was compared to a less diffused “dry-transfer,” assessing recoveries and matrix effects of 38 target compounds of emerging concern, including polar pesticides, recreational and food-related substances, pharmaceuticals, industrial additives, and ultra-violet (UV) filters. The dry-transfer procedure generally allowed better recoveries, especially for the more polar compounds, without affecting matrix effects (which remained in the range 40–130%). Along with the recovery assessment, the analysis of the non-spiked sorbent extracts revealed traces of many of the targeted emerging contaminants, quantifying perfluorooctanoic acid, UV-filters, carbamazepine, diclofenac, and triclosan. Furthermore, other compounds were found below their limits of quantitation. Ten analytes were detected only in the extracts of the dry-transferred passive samplers, highlighting the importance of applying this protocol, especially when dealing with polar compounds. This refined processing method, therefore, permits a more standardized and reproducible strategy, at the same time enlarging the set of analytes which could be detected and quantified.Graphical abstract

Predicting retention on immobilized artificial membrane chromatography: Lipophilicity-based versus lipophilicity-independent models

Retention of molecules on immobilized artificial membrane (IAM) chromatography is a key physicochemical property for predictive models of permeability across biological barriers, with applications in drug design and ecotoxicology. Currently, IAM retention is solely experimentally determined, which limits its utility for screening virtual compound libraries or for predictions of yet not synthesized molecules. The present study focuses on developing predictive models of IAM retention factors (logk_w(IAM)) for a structurally diverse set of drug compounds, scrutinizing the role of lipophilicity, experimental and calculated, as well as the contribution of additional molecular parameters, selected from a pool of physicochemical, constitutional, topological and 3D descriptors. After obtaining a data overview by principal component analysis, both multiple linear regression (MLR) and partial least squares (PLS) analyses were used to construct lipophilicity-based models and lipophilicity-independent models. Bulk, polarity and fraction of anionic species were common descriptors in all models. It was demonstrated that calculated lipophilicity values introduced additional uncertainty, depending on the software used. On the other hand, lipophilicity-independent MLR and PLS models, which relied solely on computational descriptors, showed comparable performance with lipophilicity-based models, while offering the advantage to more useful for screening large libraries in early drug discovery. The reliability of lipophilicity-independent MLR and PLS models was assessed by external validation as well as by using a blind test set. Error distribution between lipophilicity-based and lipophilicity-independent models was also investigated and found to be comparable, while it was better than the differences between experimental and calculated lipophilicity values.
